摘 要:目的:探讨贝母素乙对博莱霉素诱导的急性肺损伤大鼠TGF-β/MAPK ERK细胞信号转导通路影响。方法:大鼠麻醉后,气管内注入博莱霉素5mg/kg复制大鼠肺损伤模型,术后第2天开始每天灌胃1ml/100g体重假手术组和模型组,地塞米松0.405mg/kg组,贝母素乙5mg/kg组、贝母素乙2.5mg/kg组。28天后,麻醉并处死大鼠,肺组织固定、包埋、切片,脱蜡后,行Masson三色染色,观察肺间质炎症程度,免疫组化检测肺组织TGF-β1、MEK1/2、ERK1/2、CTGF的水平,观察贝母素乙对肺损伤大鼠TGF-β/MAPK ERK细胞信号转导通路影响。结果:贝母素乙可显著降低大鼠肺间质炎症程度,显著降低肺间质纤维化评分;贝母素乙可显著降低大鼠肺组织TGF-β1、ERK1/2、MEK1/2、CTGF水平,而与地塞米松0.405mg/kg组作用相似,贝母素乙的有效剂量为2.5mg/kg、5mg/kg。结论:贝母素乙可通过显著降低TGF-β/MAPK ERK细胞信号转导水平而减轻博莱霉素诱发的大鼠肺损伤程度。Objective: To investigate influence of peiminine on TGF- beta/MAPK ceils signal transduction pathway in acute lung injury rats induced by blcomycin. Methods: Bleomycin (5mg/kg)was instilled into the rats trachea with a microliter injector to induce acute lung injury. Rats were randomly divided into five groups: sham-operated group, control group, dexamethasone group, peiminine large group and small group. Sham operated group and control group were given Distilled water 1ml/100g weight. Dexamethasone group was given equal volume of dexam- ethasone Distilled water solution. Peiminine large group and small group were given equal volume of peiminine Distilled water solution. After daily gavage for consecutive 28 days,the rats were anesthetized and killed. Lungs were fixed, embedded, and 4urn sections were prepared. Then, the degree of pulmonary inflammation was observed by Masson three color staining and TGF-betal, MEK1/2,ERK1/2 and CTGF of the lung tissue were inspected by immunohistochemistry SABC and calculated the average optical density. Results: Peiminine can significantly re- duce pulmonary imerstitial inflammation in rats and reduce the score of pulmonary fibrosis (P 〈0.01 ). Peiminine can significantly reduce the degree of TGF-betal, MEK1/2, ERK1/2 and CTGF of the lung tissue of bleomycin -induced acute lung injury in rats. The better effective dose of peiminine is 2.5mg/kg to 5mg/kg. Conclusions: Peiminine can reduce the degree of bleomycin induced lung injury in rats by significantly reduced TGF-beta/MAPK ERK cell signal transduction pathway level.
