摘 要:目的测定丽水市一起学校病毒性胃肠炎暴发疫情诺如病毒的RNA聚合酶区(RdRp)和衣壳蛋白区(VP1)基因序列,从分子水平对病原进行分型鉴定。方法收集疫情现场现症患者肛拭子、呕吐物、各类饮用水、可疑食物留样标本总计98份,采用荧光定量PCR方法检测诺如病毒,将阳性标本通过测序引物RT-PCR扩增RdRp和VP1基因序列并测定,通过生物信息学软件进行基因分型鉴定和序列分析。结果共检出诺如病毒阳性4份,阳性检出率为4.08%(4/98),均为GⅡ基因型;RdRp和VP1序列BLAST和种系发生分型结果均显示为诺如病毒GⅡ.4亚型。结论诺如病毒优势流行株GⅡ.4亚型是本次学校病毒性胃肠炎暴发疫情的重要病原体。Objective To detect Norovirus RNA polymerase(RdRp) and shell protein area(VP1) gene sequences which outbroke in a school with viral gastroenteritis in Lishui,and molecular classification identificated of the pathogen.Methods To collect patients' anal swab,vomit,all kinds of drinking water,suspicious food retention samples in a total of 98 samples.Used fluorescence quantitative RT-PCR method to detect Norovirus,the positive specimens were amplificated RdRp and VP1 gene sequences by sequencing primers and then determinated,finally identified the genotyping and sequence through bioinformatics software.Results There were 4 positive Norovirus samples,the positive rate was 4.08%(4/98),all were GII genotype;RdRp and VP1 sequence blast and phylogenetic classification results showed that was Norovirus GII.4 subtypes.Conclusion The epidemic strains Norovirus GII.4 subtypes were the important pathogens in this school viral gastroenteritis outbreak.
