抗失眠中药复方对KM小鼠戊巴比妥钠催眠作用影响随机平行对照研究

摘　　要：[目的]观测抗失眠中药复方对KM小鼠戊巴比妥钠催眠作用影响。[方法]使用随机平行对照方法,KM小鼠140只,按体质量编号随机数字表法分为14组,空白对照组、模型组、1号药组、2号药组、3号药组、4号药组、5号药组、6号药组、7号药组、8号药组、9号药组、10号药组、百乐眠胶囊组、地西泮片组,10只/组。正交设计法筛选抗失眠中药复方制备最佳工艺路线。各组连续给药5d,末次给药后50min腹腔注射50mg·kg^(-1)戊巴比妥钠(空白组等量生理盐水),开始注射戊巴比妥钠,记录翻正反射消失时间及翻正反射恢复时间(睡眠潜伏期及睡眠持续时间)。灌胃干预40m L·kg^(-1),1次/d,连续5d,空白对照组、模型组给等容积蒸馏水。各组喂养5d(药物干预5d)。观测入睡潜伏时间、睡眠时间。[结果]与空白对照比较,模型组及各中药小复方组小鼠入睡潜伏期缩短,睡眠时间均延长(P<0.01),结果表明模型成功;与模型组比较,7号药组小鼠入睡潜伏期缩短(P<0.01),9号药组小鼠睡眠时间显著性延长(P<0.05)。表明7号药与9号药有催眠作用。[结论]抗失眠中药复方7号药、9号药与戊巴比妥钠有协同作用。[Objective] To observe the effect of anti-insomnia compound on pentobarbital sodiumhe＇s hypnotic effect. [Method] Using random parallel control method. According to body mass numbers, 140 KM mice were randomly divided into 14 groups, control group, model group, No.1 drug group, No.2 drug group, No.3 drug group, 4 drug group, No.5 drug group, No. 6 drug group, No.7 drug group,No. 8 drug group, No.9 drug group, No. 10 drug group, Bailemian capsule group and Diazepam Tablets group.There were 10 rats in each group. All rats were administered for 5 days, except for the blank group, each rat were given 50 mg ~ kg-1 pentobarbital sodium by intraperitoneal injection after 50 minutes the last administration, control group were given the same amount of saline.from the beginning of pentobarbital sodium injection, record the righting reflex time and righting reflex recovery time, sleep latency and sleep duration time. Stomach intervention 40 mL ~ kg-1, one time everyday, continuous 5 days, blank control group and model group were given equal volume of distilled water. Group 5 days （feeding drug intervention 5 days）. Observation of sleep latency and sleep time. [Results] compared with the blank control, rats＇s sleep latency and sleep time in model group and each small compound prescription of Chinese medicine group were prolonged （P〈O.01）, the results show that the model is successful; compared with the model group, No.7 medicine can shorten the sleep latency of mice （P〈0.01）; No.9 medicine can prolong the sleep time （P 〈0.05）. No.7 and No.9 medicine have hypnotic effect. [Conclusion] No.7 and No.9 small compound prescription of Chinese medicine have a certain synergistic effect with sodium.they play a hypnotic effect better.
