摘 要:将人工接种感染的小鼠腹腔细粒棘球蚴包囊90个,随机分为A^E 5组,分别用多频聚焦超声不同功率和组合方式进行超声照射.多频聚焦超声辐照后的各组包囊置于含1 00μg/mL阿苯达唑的RPMI 1640培养液中,经不同时间培养后,高效液相色谱法测定细粒棘球蚴囊内的药物质量浓度,以评价不同功率及辐照次数、辐照后含药培养液中不同培养时间对囊壁药物通透性的影响.结果显示:多频聚焦超声照射可以改变细粒棘球蚴囊壁的药物通透性.三频联合与单频相比,以及两次以上照射与单次照射相比,细粒棘球蚴囊内药物质量浓度差异均具有统计学意义.照射后包囊继续在含药培养基中培养,可进一步提高囊内药物质量浓度.本研究证实多频聚焦超声照射可使细粒棘球蚴囊壁药物通透性持续增加.多频联合超声作用的效果明显优于单频、多次照射优于单次照射.多频多次照射、适当延长包囊在药物中的培养时间能够最大程度地提高囊内药物质量浓度.Ninety of Echinococcus granulosus cysts,which came from artificially infected mice,were randomly grouped into 5 groups from A-E and were irradiated with different powers or different combinations of multifrequency focused ultrasound.All of the cysts were cultured in RPMI 1640 containing 100 μg/mL albendazole for different time after being irradiated by multi-frequency focused ultrasound.The medication concentrations in the cysts were measured by high performance liquid chromatography to evaluate all the factors,such as the different irradiation powers,the different combinations of multi-frequency focused ultrasound as well as the different time of culture in the solution containing albendazole after the irradiation on the permeability of hydatid cyst wall for medication.The results showed that multi-frequency focused ultrasound could increase permeability of Echinococcus granulosus cyst wall for medication.The medication concentrations both in the cysts irradiated by triple-frequency multi-frequency focused ultrasound compared with single-frequency irradiation and in the cysts irradiated twice by multi-frequency focused ultrasound compared with irradiated only once,showed a statistically significant difference.Prolonged culture of cysts in RPMI 1640 containing albendazole after irradiation by multi-frequency focused ultrasound could further improve the intra-cyst medication concentration.The experiment verified that multi-frequency focused ultrasound could persistently increase permeability of Echinococcus granulosus cyst wall for medication.The effect of multi-frequency irradiation and multiple-times irradiation on cysts was greater than single-frequency irradiation and only once irradiation.The multi-frequency,multiple-times irradiation and prolonged culture of cysts in medication after irradiation could increase the intra-cyst medication concentration to the maximum.
