摘 要:目的:采用紫外分光光度法和Al^(3+)显色后的比色法对超声波提取获得的新疆大叶补血草根茎中总黄酮进行含量测定,并比较两种测定结果的相关性。方法:以槲皮素为对照品,分别采用紫外分光光度法于λ=371nm直接测定和经ACl_3+KAC显色反应后于λ=436nm比色法进行测定。结果:紫外分光光度法测定新疆大叶补血草根茎中总黄酮含量为1.89%±0.0749(=6);而经显色后比色法测定结果为2.00%±0.0813(=6)。两种测定方法结果使用独立样本T检验(Independent-Samples T Test)分析,结果显示结果之间无显著性差异(=0.221)。结论:紫外分光光度法和Al_3+显色反应后的比色法均可用于新疆大叶补血草根茎中总黄酮的含量测定。Objective: Using ultraviolet spectrophotometry (UV) and colorimetric method to determine the total flavonoids content obtained from Limonium gmeIinii rhizome in Xinjiang by ultrasonic extraction process, and compare two methods results for its correlation. Methods: Using quercetin as reference substance, the total flavonoids content assay was detected by UV at 371nm and colorimetric method at 436 nm in AC13+KAC chromogenic reaction systerm, respectively. Results: The total flavonoids content was 1.89%± 0.0749,2.00% ±0.0813 (n=6)in L. gmelinii rhizome using UV and colorimetric method, respectively. The results was analysed by Independent Samples T Test. The results showed that the mean values were no significant difference with two determining methods (P=-0.221). Conclusion: The UV and colorimetric method in ACl3 + KAC ehromogenic reaction systerm could be used to detect the total flavonoids content obtained from L. gmelinii rhizome in Xinjiang.
