摘 要:目的观察道地通管汤对输卵管炎大鼠输卵管组织中转化生长因子-βⅡ型受体(TGF-βRⅡ)mRNA和Smad2 mRNA的影响。方法 68只雌性SD大鼠,随机抽取12只作为正常组,剩余大鼠采用混合菌制成输卵管炎模型,建模成功的48只大鼠分为模型组和道地通管汤低、中、高剂量组,各12只。正常组与模型组给予生理盐水灌肠,道地通管汤低、中、高剂量组分别给予不同浓度的道地通管汤灌肠[0.35 g/(100 g·d)、0.7 g/(100 g·d)、1.4 g/(100 g·d)],并于给药第15天、30天随机抽取每组6只大鼠检测其输卵管组织中TGF-βRⅡ和Smad2 mRNA的相对表达量情况。结果 (1)与正常组比,模型组给药第15、30天时TGF-βRⅡ和Smad2 mRNA的表达量均增高(P<0.05);道地通管汤各剂量组给药第30天时,差异均无统计学意义(P>0.05)。(2)与模型组比,给药第15、30天时道地通管汤中、高剂量组TGF-βRⅡ和Smad2mRNA的表达量均降低(P<0.05),给药第15天时道地通管汤低剂量组TGF-βRⅡ的表达量也降低(P<0.05)。(3)道地通管汤各剂量组中TGF-βRⅡ和Smad2 mRNA的表达量呈低剂量组>中剂量组>高剂量组,给药第30天时道地通管汤低剂量组与道地通管汤高剂量组的TGF-βRⅡ表达量比较,差异有统计学意义(P<0.05),其余组间比较,差异无统计学意义(P>0.05)。(4)道地通管汤各剂量组中,给药第30天时TGF-βRⅡ和Smad2 mRNA的表达量均低于给药第15天,其中道地通管汤各剂量组中Smad2 mRNA的表达量组内比较,差异有统计学意义(P<0.05)。结论道地通管汤能抑制输卵管炎性大鼠输卵管组织中TGF-βRⅡ和Smad2的mRNA表达;道地通管汤给药30 d作用效果较好,且道地通管汤中剂量与高剂量作用效果相近。Objective To observe the effect of Daodi Tongguan decoction on the expressions of transforming growth factor-β receptor Ⅱ ( TGF-βRⅡ) mRNA and Smad2 mRNA in fallopian tube tissues of rats with salpingitis. Methods Twelve of 68 female SD rats were selected randomly as the normal group, and salpingitis models were established using mixed bacteria in the rest rats. The 48 successful rat models were divided into model group ,low-,medium- and high-dose Daodi Tongguan decoction groups ,with 12 rats in each group. The rats in the normal group and model group were given normal saline by enemata, while the rats in the low-, medium- and high-Daodi Tongguan decoction groups were given Daodi Tongguan decoction with the concentrations of 0.35 g/( 100 g · d),0.7 g/( 100 g · d) and 1.4 g/( 100 g· d) by enemata respectively. On the 15th and 30th day after administration,six rats were randomly selected from each group to detect the relative expressions of TGF-βRⅡ and Smad2 mRNAs in their fallopian tube tissues. Results ① Compared to the normal group,the expression quantities of TGF-βRⅡ and Smad2 mRNAs in the model group increased on the 15th and 30th day after administration (P 〈0. 05) ; on the 30th day after administration,no significant differences in the expression quantities of TGF-βRⅡ and Smad2 rnRNAs were found among allDaodi Tongguan decoction groups (P 〉 0.05 ). ② Compared to the model group ,the expression quantities of TGF-βRⅡ and Smad2 mRNAs in the medium- and high-dose Daodi Tonggnan decoction groups decreased on the 15th and 30th day after administration(P 〈 0.05), and the expression quantity of TGF-βRⅡ in the low-dose Daodi Tongguan decoction group also decreased on the 15th day after administration (P 〈 0. 05). ③ Among all Daodi Tongguan decoction groups,the expression quantities of TGF-βRⅡand Smad2 mRNAs deceased in the order of low-,medium- and high-dose Daodi Tongguan decoction groups.
