摘 要:目的 观察给予正常成年大鼠脑内外源性N-甲基-D-天冬氨酸(NMDA)对其空间学习记忆的影响及海马区NMDA受体活性的改变.方法 选取正常成年雌性Wister大鼠90只,随机分成6组,正常大鼠组(组1)、手术对照组(组2)、空白膜片组(组3)、10^-5 mmol/L浓度组(组4)、10^-4 mmoL/L浓度组(组5)及10^-3 mmol/L浓度组(组6).第1组为正常大鼠,不予以任何处理;第2组大鼠仅行开颅手术;其余4组大鼠开颅手术后分别在大脑皮层表面置入一空白膜片或上述3种浓度的NMDA缓释膜片.手术后3周,利用Morris水迷宫测试评价各组大鼠空间学习记忆能力,并用放射标记受体测定方法测定各组大鼠海马区NMDA受体活性.结果 1-6组大鼠Morris水迷宫测试的潜伏期分别为:(38.3±25.6)、(30.9±21.9)、(28.1±21.1)、(24.4±17.1)、(25.3 ±21.8)、(24.0±15.5)s,各组潜伏期之间差异无统计学意义(P>0.05).各组大鼠海马区的NMDA受体活性分别为:(45313.57 ± 11851.33、43 793.84±6 835.53、47 962.06±5019.62、42 699.19±5 778.45、45 521.03 ±9 924.43、51 408.68±15 041.08) fmol/(mg·prot).各组间测定结果差异无统计学意义(P>0.05),但随着所植入的缓释膜的NMDA浓度增加,大鼠海马区NMDA受体活性有增高的趋势.结论 不同浓度缓慢释放NMDA的缓释膜片没能对正常大鼠的空间学习记忆能力产生显著影响.使用的NMDA浓度对正常大鼠相对安全,无明显毒性反应。Objective To observe the effect of exogenous N-methyl-D-aspartate (NMDA) on the spatial learning and memory ability associated with NMDA receptors activity within hippocampus of normal adult rats.Methods Ninety female Wister rats were randomly divided into 6 groups:normal rats (group 1),rats operated without planting film (group 2),rats operated with films without NMDA (group 3),rats operated with 10^-5 mmol/L (group 4),10^-4 mmol/L (group 5) and 10^-3 mmol/L (group 6) NMDA films.Rats in group 1 were not given any interference.Rats in group 2 were operated without planting film.Rats in other four groups were operated,followed by planting films without containing NMDA or with slow-release films containing different NMDA concentrations on the surface of cortex respectively.Three weeks after operation,we evaluated the spatial learning and memory ability of the rats in each group by Morris water maze test,then used radiolabeled receptor assay to measure the NMDA receptors activity in hippocampus region.Results The latency of the 6 groups in Morris water maze test were (38.3 ±25.6),(30.9 ± 21.9),(28.1 ± 21.1),(24.4 ± 17.1),(25.3 ± 21.8) and (24.0 ± 15.5) s,respectively,and the results showed that there was no statistically significant difference among all groups (P 〉 0.05).The NMDA receptors activity in hippocampus region was (45 313.57 ± 11 851.33),(43 793.84±6835.53),(47962.06± 5019.62),(42699.19± 5778.45),(45521.03± 9924.43),and (51 408.68 ± 15 041.08) fmol/(mg·prot),respectively,and the results showed that there was no statistically significant difference among all groups (P 〉 0.05).Conclusion The slow-release films with different NMDA concentrations failed to have a significant effect on the spatial learning and memory ability of normal rats.The different NMDA concentrations were relatively safe for normal rats,and there was no obvious toxic reaction.
