摘  要:以燕子掌叶片为试材,采用植物组织培养方法,研究了消毒时间、不同浓度2,4-D、6-BA、NAA对燕子掌再生的影响。结果表明:叶片用70%~75%的酒精浸泡10~15s,再用0.1%HgCl2消毒12~15min,具有较低污染率和较高的存活率;用MS+1.5mg·L^(-1) 6-BA+0.5mg·L^(-1)2,4-D进行不定芽诱导,平均可以产生13.7个不定芽;不定芽在MS+6-BA 1.5mg·L^(-1)+NAA0.1mg·L^(-1)+GA30.5mg·L^(-1)中增殖倍数为11.0;在1/2MS+IBA 0.5mg·L^(-1)或MS+IBA0.5mg·L^(-1)培养基中平均能产生9~11条根。Taking Crassula arborescens leaves as materials,using plant tissue culture method,the effects of sterilizing time and different concentration of 2,4-D,6-BA,NAA on regeneration of Crassulaarborescens were studied.The results showed that soaking the leaf in 70%-75%alcohol for 10^(-1)5 seconds,then 0.1% HgCl2 disinfection 12^(-1)5 minutes,the callus had low contamination rate and high survival rate.MS+1.5 mg·L^(-1) 6-BA+0.5 mg·L^(-1) 2,4-D was optimum medium for inducing adventitious buds with an average of 13.7adventitious buds;adventitious buds in MS+6-BA 1.5mg·L^(-1)+NAA 0.1mg·L^(-1)+GA30.5mg·L^(-1),the proliferation coefficient was 11.0;multiple shoots could produce an average of 9^(-1)1 roots in the 1/2MS+IBA 0.5mg·L^(-1) or MS+IBA 0.5mg·L^(-1) medium.
