凌霄花总黄酮对脑缺血大鼠脑组织中炎性因子及趋化因子的影响

摘　　要：目的:通过观察凌霄花总黄酮对脑缺血大鼠脑组织中炎性因子和趋化因子的影响,研究其对脑缺血急性期炎性反应的阻抑作用。方法:复制大脑中动脉阻塞大鼠模型,缺血2h,再灌注22h后,取脑,制备10%脑匀浆,ELISA法检测脑组织中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、单核细胞趋化因子-1(MCP-1)、激活调节细胞因子(RANTES)的水平,分光光度法检测骨髓过氧化物酶(MPO)活力。结果:与假手术组比较,模型组大鼠脑组织中IL-6、IL-1β、MCP-1、RANTES、MPO水平和活力明显升高(P<0.01);与模型组比较,凌霄花总黄酮能明显降低IL-6、IL-1β、MCP-1、RANTES、MPO水平和活力(P<0.01,P<0.05)。结论:凌霄花总黄酮通过抑制脑缺血大鼠脑组织中炎性因子和趋化因子的水平,对脑缺血急性期的炎性损伤有一定的阻抑作用,从而产生脑保护作用。Objective：To observe the influence of total flavonoids of Campsis grandil ora on inflammation cytokines and chemokines in brain tissue of rats with cerebral ischemic,and study its inhibition on inflammatory response during the acute phase of cerebral ischemia.Methods：Rat models of transient middle cerebral artery occlusion were established.After 2h of ischemia and 22 h of reperfusion,the brain tissues were obtained to prepare 10% brain homogenate.The levels of IL-6,IL-1β,MCP-1,RANTES in brain tissue of rats were tested by using ELISA method,and the activity of MPO was tested by using spectrophotometry.Results：Compared with sham operation group,the levels of IL-6,IL-1β,MCP-1,RANTES,MPO,and activity of MPO in brain tissue of rats in model group increased significantly（P〈0.01）.Compared with model group,total flavonoids of Campsis grandil ora could decrease the levels and activity of IL-6,IL-1β,MCP-1,RANTES,and MPOsignificantly（P〈0.01,P〈0.05）.Conclusion：Total flavonoids of Campsis Grandil oracould inhibit the inflammatory injury during acute phase of cerebral ischemia and protect brain tissue through inhibiting the levels of inflammation cytokines and chemokines in brain tissue of rats with cerebral ischemic.
