益气化瘀清热方及其拆方对大鼠肾系膜细胞IκB-α mRNA及蛋白信号通路的影响

摘　　要：目的:对益气化瘀清热方进行拆方,观察各类含药血清对脂多糖(LPS)诱导下体外培养的大鼠肾小球系膜细胞(Ms C)表达IκB-αm RNA及其蛋白的影响,探讨不同中药的作用强度以及益气化瘀清热方治疗肾小球肾炎的作用机制。方法:采用RT-PCR法检测各组含药血清对Ms C表达IκB-αm RNA的影响,Wersten blot法检测各组含药血清对大鼠Ms C表达IκB-α蛋白的影响。结果:各组含药血清均能上调体外培养的Ms C表达IκB-αm RNA及蛋白,清热组作用大于化瘀组、益气组,其中益气组作用最弱,三者之间的差异均有统计学意义(P<0.05);与雷公藤多苷组比较复方组作用较强(P<0.05),清热组无显著性差异。结论:益气化瘀清热方及其拆方均能上调体外培养的大鼠Ms C表达IκB-αm RNA及其蛋白,复方组作用明显优于其他组,清热组、雷公藤多苷组次之,益气组最弱。Objective：To study the mechanism of Yiqi Huayu Qingre Formula in treating glomerulonephritis and the effect of different herbs in the decoction by observing the effects of serums containing different medicine on the expression of IκB-αm RNA and the protein in primary mesangial cells（Ms C） of rats cultured in vitro and induced by lipopolysaccharide（LPS）.Methods：Effects of serums containing different medicine on Ms C＇s expression of IκB-α m RNA and IκB-α protein were detected with the methods of RT-PCR and Western Blot respectively.Results：All the groups of medicine can up-regulate Ms C＇s expression of IκB-α m RNA and the protein,among which the effect of the Qingre group was stronger than that of Huayu group and Yiqi group with Yiqi group as the weakest.The differences among the three groups were statistically significant（P〈0.05）.Compared with TW group,the effects of the compound group was better（P〈0.05）.Conclusion：Yiqi Huayu Qingre Formula,its disassembled formulas and TW can all inhibit the expression of IκB-α m RNA and protein in Ms C of rats.The effects of the compound group is superior to that of other groups.The Yiqi group has the weakest effect.
