摘 要:目的:建立在同一色谱条件下同时测定丹参多组分复合微乳中丹参酮ⅡA和丹酚酸B的HPLC含量测定方法。方法:采用Kromasil C18(250mm×4.6mm,5μm)色谱柱,以乙腈-0.4%磷酸溶液(0-20min,10-45∶90-55;20-21min,45-80∶55-20;21-37min,80∶20;37-38min,80-10∶20-90,38-43min,10∶90)为流动相,流速1.2m L/min,丹参酮ⅡA和丹酚酸B的检测波长分别为270、286nm,进样量10μL。结果:丹参酮ⅡA和丹酚酸B分别在0.3-64μg/m L和10-210μg/m L范围内线性关系良好,加样回收率RSD均小于3%,以标示量计算的平均含量分别为(957.17±1.04)μg/g和(1 315.08±15.66)μg/g。结论:本方法简便、稳定、用时较少。该色谱条件下,各成分分离度良好,可用于丹参复合微乳的含量测定。Objective:To establish an HPLC method for the simultaneous determination of the contents of tanshinone ⅡA and salvianolic acid B in multicomponent compound microemulsion of Salvia miltiorrhiza Bge.under the same chromatographic condition.Methods:The determination was performed on a Kromasil C18 chromatographic column(250mm×4.6mm,5μm) with a mobile phase of acetonitrile-0.4% phosphate solution(0-20 min,10-45∶90-55;20-21 min,45-80∶55-20;21-37 min,80∶20;37-38 min,80-10∶20-90,38-43 min,10∶90).The flow rate was 1.2m L/min,the detection wavelength of tanshinone II A and salvianolic acid B were 270 nm and 286 nm respectively.The injection volume was 10μL.Results:Good linear relationship between tanshinone II A and salvianolic acid B was achieved in the concentration of 0.3-64μg/m L and 10-210μg/m L respectively.The recovery RSD was less than 3%.The average contents of tanshinone II A and salvianolic acid B by indicated amount were(957.17±1.04) μg/g and(1 315.08±15.66) μg/g respectively.Conclusion:This method is simple,stable and time-saving,with a good resolution of each composition under this chromatographic condition.The method can be used for content determination of the compound microemulsion of Salvia miltiorrhiza Bge..
