葛根素对大鼠急性脊髓损伤影响的实验研究

摘  要:目的探讨葛根素对大鼠急性脊髓损伤的保护作用。方法:63只雄性Wistar大鼠,按随机化表法分为假手术组、损伤组、葛根素组,每组21只,采用改良的A1len’s撞击法制成脊髓损伤模型。假手术组仅暴露,不损伤;损伤组制大鼠脊髓损伤模型;葛根素组在损伤造模后,于1、3、6、24、48h分别腹腔注射葛根素素(50mg/kg),余两组仅注射同量的生理盐水。结果:损伤组和葛根素组在伤后24、48、72h脊髓组织含水量均高于假手术组:且葛根素组脊髓组织含水量明显低于损伤组。与假手术组比较,损伤组和葛根素组在伤后24、48、72hSOD活性均显著下降,NDA含量均显著上升;而葛根素组与损伤组比较,SOD活性的下降及MDA含量的上升均显著减缓。在伤后14d电镜下观察,损伤组神经纤维仍损伤明显:而葛根素组神经纤维修复趋于正常。结论:葛根素对大鼠急性脊髓损伤有保护作用,其作用机制可能部分通过清除自由基,抑制脂质过氧化,从而减轻神经细胞的继发性损伤来发挥其保护作用。Objective To investigate the protective effect of puerarin on acute spinal cord injury in rats. Methods:Totally 03 male Wistar rats were divided into 3 groups by random table method, each group with 21 rats: Sham operation group only exposed but no injured spinal cord, injury group received spinal cord injury by Allen's method, puerarin group in injury models were made and were intraperitoneal injection puerarin (50mg/kg), in the other grouPs only injection amounts of saline in lh, 3h, 6h, 24h, 48h. Results:The contents of water in the injured group and the puerarin group were higher than those in the sham operation group at 24h, 48h and 72h after injury, and the water content in the spinal cord of the puerarin group was significantly lower than that in the injury group. Compared with the sham group, injury group and puerarin group after injury in 24h, 48 h and 72 h of SOD activity were significantly decreased, MDA content increased significantly; and puerarin group and injury group compared the SOD activity decreased and MDA content increased significantly slowed. Observation under electron microscope at 14d after injury, injury group of nerve fibers is still obvious injury and repair of nerve fibers; puerarin group tended to be normal. Conelusions:Puerarin has a protective effect on acute spinal cord injury in rats, its mechanism may in part by scavenging free radicals and inhibiting lipid peroxidalion, reducing secondary injury of nerve ceils to exert its protective effect.
