益精方对腺嘌呤法大鼠睾丸支持细胞中紧密连接蛋白表达的实验研究  被引量:3

摘  要:目的:观察益精方对腺嘌呤法不育症大鼠睾丸支持细胞中紧密连接蛋白表达的影响。方法:选用48只健康的SPF级Wistar雄性大鼠,2月龄,体质量(180±20)g,按随机数字表法,平均分为空白组,模型组,复方玄驹组,益精方组,每组12只。除空白组外,其余各组均以腺嘌呤1 m L/(100g·d)的剂量连续灌胃12 d;从第13天开始,空白组及模型组用等量生理盐水灌胃,复方玄驹组以复方玄驹胶囊水溶液灌胃,益精方组以益精方汤剂灌胃,每天1次,连续20 d。实验结束后处死大鼠,摘取睾丸和附睾,采用计算机精子分析系统计数精子密度和精子活动率;采用免疫组化技术测定大鼠睾丸支持细胞中紧密连接蛋白claudin的表达。结果:与空白组比较,模型组中精子活动率、精子密度、claudin蛋白表达均降低,差异均具有统计学意义(P<0.01)。与模型组比较,复方玄驹组精子活动率、精子密度均升高,差异具有统计学意义(P<0.05),claudin蛋白表达升高不明显,差异无统计学意义(P>0.05),益精方组精子活动率、精子密度、claudin蛋白表达均升高,差异具有统计学意义(P<0.05)。结论:益精方能够提高腺嘌呤诱导的肾阳虚模型大鼠的精子密度和精子活动率水平;同时使claudin蛋白的表达增多。Objective: To observe the influence of Yijingfang on the expression of tight junction protein in testicular sertoli cells of rats by adenine method. Methods : Select 48 healthy male Wistar rats of SPF grade which were 2 - month old with weight( 180 _+ 20)g. The rats were randomly divided into blank group ,model group, compound Xuanju group ,Yi- jingfang group,12 rats in each group. Except the blank group,the other groups were given adenine 1 mL/( 100 g ~ d) by intragastric administration for 12 days. From the thirteenth day on ,the blank group and model group were given the nor- mal saline and compound Xuanju group was treated with water solution of compound Xuanju Capsule and Yijingfang Group with Yijingfang intragastrically, once a day for 20 consecutive days. At the end of the experiment, the rats were sacrificed and the testis and epididymis were taken out. By using the computer sperm analysis system, we counted the rate of sperm density and sperm activity and by immunohistochemical technique, determined the expression of the protein claudin in rats' testis sertoli cells. Results : Compared with the blank group, model group' s sperm activity, sperm density and clau- din protein expression were reduced and the differences were statistically significant ( P 〈 O. 01 ). Compared with model group, Xuanju group' s sperm activity and sperm density were higher and the difference was statistically significant ( P 〈 0.05 ) but elaudin protein expression was not obvious and there was no statistically significant difference ( P 〉 0.05 ). Yi- jingfang group' s sperm activity, sperm density and claudin protein expression were higher with statistically significant difference ( P 〈0. 05 ). Conclusion : Yijingfang can improve the levels of sperm density and sperm activity in kidney Yang deficiency model rats induced by adenine, while the expression of claudin protein increased.
