淫羊藿苷对氧糖剥夺PC12细胞活力和细胞形态的影响

摘　　要：目的:观察淫羊藿苷对PC12细胞的毒性作用以及不同给药方法、给药剂量对氧糖剥夺PC12细胞活力的影响。方法:将不同浓度的淫羊藿苷与PC12细胞作用24 h后,用MTT法检测细胞活力。用预给药1 h,氧糖剥夺期间给药2 h,预给药1 h+氧糖剥夺期间给药2 h方法给药,细胞复氧复糖24 h后,用MTT法检测细胞活力,预给药1 h+氧糖剥夺期间给药2 h组在造模结束后用倒置显微镜观察细胞形态。结果:PC12细胞用10^(-8)~10^(-4)mol/L淫羊藿苷孵育24 h后,细胞活力与正常组比较均显著下降(P<0.01);10^(-8)~10^(-5)mol/L淫羊藿苷预给药1 h均使模型细胞的活力显著升高(P<0.01);10^(-8)~10^(-4)mol/L淫羊藿苷氧糖剥夺期间给药2 h均使模型细胞的活力显著下降(P<0.01);10^(-8)~10^(-5)mol/L预给药1 h+氧糖剥夺期间给药2 h均使模型细胞的活力显著升高(P<0.01),使模型细胞数量增加,细胞形态改善。结论:淫羊藿苷与细胞的作用时间不宜过长,预给药1h+氧糖剥夺期间给药2 h对氧糖剥夺-复氧PC12细胞能起到较好的保护作用。Objective：To investigate the toxicity of PC12 cells and the effects of different methods of administration, dosage on cell viability in PC12 cells injured by oxygen and glucose deprivation （OGD）. Methods ：Treatment with differ- ent codcentrations of icariin for 24 h,the viability of PC12 cells was measured by using MTT assay. Pre -treatment with icariin for 1 h, treatment with icariin for 2 h during OGD, and pre - treatment with icariin for I h and lasting for 2 h during OGD,after 24 h reoxygenation, the cell viability was measured by MTT assay. After modeling, cell morphology in the group of pre - treatment with icariin for 1 h and lasting for 2 h was observed by using an inverted microscope. Results ： Af- ter incubation with 10-8 · 10-4 mol/L icariin for 24 h,the cell viability of PC12 ceils was significantly decreased com- pared with normal group,P 〈0.01. After pre - treatment with 10 -s · 10 -5 mol/L icariin for 1 h,the cell viability of the model cells was significantly increased （ P 〈 0.01 ）. After treatment with 10 -s · 10 -4 mol/L icariin for 2 h during OGD, the cell viability of the model cells was significantly decreased （P 〈 0.01 ）. After pre -treatment with 10-s· 10-5 mol/ L icariin for 1 h and lasting for 2 h,the cell viability of the model cells was significantly increased （ P 〈 0.01 ）. The num- ber of model cells was increased and cellular morphology was improved. Conclusion ： The incubation time of icariin was not too long. Pre - treatment with 10 -s · 10 -s mol/L icariin for 1 h and lasting for 2 h can play a better protection in PC12 cells following OGD/R injury.
