摘 要:目的:探讨白念珠菌Hogl基因DNA、mRNA、蛋白的潜在生物学性质。方法:采用生物信息学方法对白念珠菌Hogl基因DNA、mRNA、蛋白序列进行分析。结果:白念珠菌Hogl基因启动子可能位于其基因编码区起始位点前654~704bp,转录起始位点为编码区起始位点前663bp,Hogl基因并无内含子。Hogl基因mRNA中未发现核糖体结合位点等功能性RNA序列,RNA二级结构分析显示,HoglmRNA形成稳定的回文结构。蛋白质的理化性质分析显示,Hogl蛋白稳定系数为35.67。TargetP1.1和SignalP4.1预测结果显示,Hogl基因不存在线粒体定位序列、分泌信号肽及跨膜区域。Motif分析显示Hogl具有MAPK活性,其活性位点可能为第189—181个氨基酸(TRY)。Hogl蛋白序列在各物种间呈渐进式进化、较为保守,尤其在MAPK功能域附近。结论:白念珠菌Hogl基因的DNA和mRNA序列无常见功能域,可能主通过其蛋白质发挥作用。Hogl蛋白是MAPK家族成员,其MAPK活性位点(第189—181个氨基酸TRY)或为潜在的药物靶点。Objective :To explore the biological characters of the DNA, mRNA and protein se- quence of Hogl gene from the Candida albicans. Methods: The bioinformatical analysis of DNA, mRNA and protein sequence of Hogl gene from the Candida albicans was conducted. Results: The promoter of Hogl gene was probably located 654-704bp before the starting site of the coding region. The transcription start site of Hogl was 663bp prior to the starting site of the coding region. Hogl had no introns. The Hogl mRNA was absent of functional RNA sequences, including the ri-bosome binding sites. RNA structure analysis showed that Hogl mRNA forms a stable palindrome. The analysis of the physical and chemical properties showed that the stabilizing factor of Hogl pro- tein was 35.67. TargetP 1.1 and SignalP 4.1 predicted that Hogl gene was lack of mitochondrial localization sequence, secretion signal peptide and transmembrane domain. Motif analysis revealed Hogl had MAPK activity and the active site might be the 189-181rst amino acids (TRY). Hogl protein was gradually evoluted and conservative among species, especially in the MAPK domain. Conclusion: The DNA and mRNA sequences of Hogl gene from Candida albicans have no com- mon functional domains, which indicated that Hogl might play its role with protein. Hogl gene may be a marker of molecular evolution and its 189-181rst amino acid (TRY) might be a potential drug target.
