凋膜止崩方对大鼠子宫内膜增生症组织中MMP-1及TIMP-1mRNA表达的影响

摘　　要：目的探讨凋膜止崩方对大鼠子宫内膜增生症子宫内膜组织中MMP-1及TIMP-1mRNA表达的影响,为一种临床治疗无排卵型功血(ADUB)的新方法 ---中药靶向干预子宫内膜增生症提供理论依据。方法应用RT--PCR技术从基因水平检测凋膜止崩方对去势雌性大鼠增生症子宫内膜组织中MMP-1及其抑制因子TIMP-1 mRNA表达的影响。结果用药组子宫内膜组织中MMP-1mRNA的表达均显著低于正常空白组,而TIMP-1mRNA的表达均显著高于正常空白组(P<0.05)。结论凋膜止崩方对子宫内膜增生症组织中MMP-1及其抑制因子TIMP-1mRNA有显著影响,抑制MMP-1mRNA的表达,而促进TIMP-1mRNA的表达,破坏增生过长的子宫内膜,达到彻底治愈功血的目的,替代宫腔镜下子宫内膜电切术(TCRE),克服了手术的部分副作用,值得临床推广应用。Objective ： To study the effect of Diaomozhibeng Prescription on MMP - 1 and TIMP - 1 mRNA expression of endometrial hy- perplasia tissues in rats and to provide a theoretical basis for anovulatory dysfunctional uterine bleeding（ADUB） method,i, e. TCM targeted intervention endometrial hyperplasia. Methods： RT - PCR technique was used to detect genetically the effect of Diaomozhibeng Prescription on MMP - 1 and TIMP - 1 mRNA expression of endometrial hyperplasia tissues in ovariectomized rats. Results ： The expression of MMP - 1 mRNA endometrial tissue of the treatment group was significantly lower than that of the normal control group, but the expression of TIMP - l mRNA was significantly higher than that of the normal control group（ P 〈 O. 05 ）. Conclusion ：Diaomozhibeng Prescription has a significant effect on MMP- 1 and TIMP- 1 mRNA inhibitors, inhibit the expression of MMP- 1 mRNA, but promote the expression of TIMP- 1 mRNA to undermine uterine hyperplasia so as to achieve the goal of curing DUB and replace transcervical resection of endometrium（TCRE）to o- vercome some of side effects of surgery,worthy of clinical application.
