摘 要:目的:观察人参皂甙Rg1对大鼠坐骨神经损伤后的修复作用,寻求最佳剂量。方法:健康成年SD大鼠90只,采用离断左侧坐骨神经的方法建立周围神经损伤模型,建模后随机分为6组(实验组2 mg组、4 mg组、8 mg组、12 mg组,甲钴胺组和空白对照组),每组15只。实验组大鼠腹腔注射人参皂甙Rg1 2 mg/kg、4 mg/kg、8 mg/kg和12 mg/kg,甲钴胺组大鼠给予甲钴胺(100μg/kg),空白对照组给予等量生理盐水。给药8周后行坐骨神经功能指数(sciatic functional index,SFI)和神经传导速度检测。结果:8周后各实验组和甲钴胺组大鼠SFI、神经传导速度均优于空白对照组(P<0.05);实验各组之间比较,8 mg组优于其他各剂量组(P<0.05)。结论:人参皂甙Rg1接近8 mg/kg剂量治疗大鼠坐骨神经损伤的作用较好。Objective: To study the repairing effects of ginsenoside Rg1 on rat sciatic nerve after injury and to seek the optimal dose. Methods:90 healthy adult SD rats,with left sciatic nerve apocoped as model for treatment of peripheral nerve injury,were randomly divided into 6 groups( experiment group 2 mg、4 mg、8 mg、12 mg,mecobalamine group and blank control group),15 rats in each group. Ginsenoside Rg1 2 mg/kg,4 mg / kg,8 mg / kg and 12 mg / kg were injected into abdominal cavity in the experiment groups. Mecobalamine( 100 μg / kg) was given in the mecobalamine group and the rats in the blank control group were injected Sodium Chloride at the same dosage. The sciatic function index( SFI)and nerve conduction velocity were detected after 8 weeks administration. Results: SFI and nerve conduction velocity in the experiment groups and the mecobalamine group were better than those in the blank control group( P〈0. 05); SFI and nerve conduction velocity in the 8 mg group were significantly better than those in the other experiment groups( P〈0. 05). Conclusion: Ginsenoside Rg1 8 mg / kg is close to the optimal dose in the treatment of sciatic nerve injury.
