摘 要:为探讨印防己毒素(PTX)阻断抑制性自发和诱发的神经递质释放浓度是否存在差异,以及这些差异是否具有脑区特异性,采用鼠脑皮层神经细胞和海马神经细胞作为实验材料,在外液中加入不同浓度的PTX,分别记录了m IPSC的频率和e IPSC的幅值.结果发现:PTX作用于自发性神经递质释放的半抑制浓度(IC50)显著小于诱发性神经递质释放的,说明自发神经递质释放对于PTX的作用更为敏感.但是皮层细胞和海马细胞之间各项参数的差异较小,说明PTX在各脑区中的作用浓度相似.因此,PTX阻断抑制性神经递质自发释放和诱发释放可能有不同的机制,但并不具有脑区特异性.To investigate if there did exist the differential regulation between inhibitory spontaneous and evoked neurotransmitter release blocked by different concentration of picrotoxin (PTX) and to explore whether the difference had the brain regions specificity, neuronal cultures were obtained from mouse cortex and hippocampus, respectively. We monitored spontaneous mIPSCs and elPSC in different extracellular concentration of PTX, respectively, and observed that the half maximal inhibitory concentration (IC50) of the mIPSCs was significantly smaller than that of the elPSC, indicating that the spontaneous release was more sensitive when PTX applied. But no obvious difference between cortical and hippocampal cells was revealed in our experiments. Therefore, our results suggested that PTX had differential regulating mechanisms between inhibitory spontaneous and evoked neurotransmitter release, but without brain regions specificity.
