益胃汤对子宫内膜异位症模型大鼠Th_1/Th_2漂移的影响

摘　　要：目的:观察子宫内膜异位症模型大鼠血清IL-4,INF-γ水平和血浆IL-10的变化,研究益胃汤对子宫内膜异位症(Endometriosis,EMs)模型大鼠外周血Th1/Th2平衡的干预作用。方法:选择40只造模成功的自体移植EMs模型大鼠随机分成模型对照组、丹那唑组(0.02 g/kg)、益胃汤高(27.00 g/kg)、中(13.5 g/kg)、低(6.75 g/kg)三剂量组,并取8只建立假手术组,药物组和假手术组均采用灌胃给药方式给药,治疗组给予对应药物,给药体积10 m L/kg,一次/d,假手术组给予等体积蒸馏水,药物连续干预4 w,4 w后取异位子宫内膜观察其病理形态学以及观察大鼠血清IL-4,INF-γ水平和血浆IL-10,计算IL-4/INF-γ和IL-10/INF-γ比值并分析其变化。结果:与假手术组比较,模型对照组大鼠子宫内膜纤维组织增生,内膜呈乳头状增生,腺体呈囊性扩张明显,可见腺体及内膜间质浸及肌层,内膜间质变小;IL-10/INF-γ显著升高(P<0.05)。与模型对照组比较,益胃汤各给药组大鼠子宫内膜病理形态学均显著改善,血清INF-γ显著增高(P<0.05),IL-4/INF-γ和IL-10/INF-γ比值均明显变小(P<0.05)。结论:益胃汤改善外周血炎症因子浓度,减轻临床症状,其作用机制之一EMs可与其调节Th1/Th2失衡有关。Objective： Observing the changes of of IL-4, INF-y in serum and IL-10 in plasma in the Endometriosis （EMs） model ratsand to research intervention effect of Yiwei Tang on the balanced of Thl/Th2 in the rats. Methods： Choose 40 successful autograft EMs mode rats and divided them into five groups stochastically： model control group, Danazol group （0. 02 g/kg）, YWT high-dose group （27. 00 g/kg）, YWT mid-dose group （ 13.50 g/kg）, YWT low-dose group （6. 75 g/kg） and elected anther 8 rats to establish sham group, each groups were treated by lavage with theirs drugs respectively, 10ml//kg, 1 times/day, the sham group treated with isometric distilled water and intervened them 4w. Made pathology slices with the ectopic endometrias after treated them 4 weeks and detecting the concentration of IL-4, INF-y in serum and IL-10 in plasma by ELISA and calculated the value of IL-4/INF-y and IL-10/INF-y. Results： Compared with sham group, There are fibrosis of endometria, papillary hyperplasia, evidently cystic dilated of glands, glands and stroma infiltrating into the layer of muscle, the cutdown of stroma of endometria in the model group; and IL-10/INF-y is increase distinguished （P 〈 0. 05） . Compared with model group, Pathological and morphology are ameliorated observably in Yiwei tang groups, INF-y of serum level is increase observably （ P 〈 0. 05 ）, the value of IL-4/INF-y and IL-10/INF-y are reduce markedly （ P 〈 0. 05 ） . Conclusion： Yiwei Tang can alter the concentration of inflammatory factors, reduces the elinlcal symptom, a potent machine of this changes is to regulate the imbalanced of Th1/Th2.
