摘  要:目的 探讨不同吸烟周龄小鼠的精子活率和睾丸组织外周致密纤维1(ODF1)mRNA的表达。方法 6~8周龄的C57BL/6J健康雄性小鼠分别给予吸烟2周、4周和6周处理,ELISA试剂盒检测血清可替宁含量,血球计数板计数精子活率,实时荧光定量PCR检测ODF1mRNA表达水平的改变。结果 经过2周、4周和6周的烟雾处理后,小鼠血清中可替宁含量明显高于不吸烟组,差异均有统计学意义(P〈0.05)。吸烟6周后,小鼠血清中可替宁含量可达(10.76±2.19)ng/ml。此外,吸烟6周小鼠的睾丸组织ODF1mRNA表达量明显低于对照组(P〈0.05)。经过烟雾处理后,小鼠精子活率并没有发生显著改变。结论 本实验成功建立吸烟小鼠模型,可以为其他疾病的研究提供吸烟小鼠模型。其次,吸烟可能通过影响ODF1基因的表达而作用于精子的发生阶段,从而影响男性生殖系统。Objective To explore sperm motility rate of mice with cigarette smoking history for different weeks and expression of outer- dense fiber 1 (ODF1) mRNA in testicular tissue. Methods The healthy male C57BL/6J mice of 6-8 weeks were treated with cigarette smoking for 2, 4 and 6 weeks, ELISA kit was used to detect serum cotinine levels, hemocytometer was used to count sperm motility rate, re- al-time fluorescent quantitative PCR was used to detect the change of ODFI mRNA expression level. Results After treated for 2, 4 and 6 weeks, the levels of serum cotinine in cigarette smoking group were statistically significantly higher than those in non-cigarette smoking group ( P〈0. 05 ) . After treated for 6 weeks, the level of serum cotinine was ( 10.76±2. 19) ng/ml. The expression level of ODF1 mRNA in tes- ticular tissue of mice treated for 6 weeks was statistically significantly lower than that in control group (P〈0. 05) . After treatment, sperm motility rate did not change significantly. Conclusion The study has successfully established the mouse model of cigarette smoking and may provide a mouse model for study of other diseases. Additionally, cigarette smoking may affect spermatogenesis by affecting the expression of ODF1 mRNA, which may have impact on male infertility.
