茜草素联合抗结核西药对耐多药结核分枝杆菌体外抗菌活性研究

摘　　要：目的评价茜草素对耐多药结核分枝杆菌的体外抗菌活性作用。方法采用人型结核分枝杆菌标准菌株(H37RV)、耐多药结核菌株(MDR-TB)分别接种于含有不同浓度茜草素单用、联合抗结核西药、对照组的结核菌培养基,观察结核分枝杆菌的生长情况,评价茜草素体外抑菌效果。结果模型组菌落生长情况正常,结核分枝杆菌(H37RV)培养终止期茜草素(80μg/ml)单用组、利福平(5mg/ml)组未见菌落生长;茜草素(80μg/ml)单用组培养基恢复培养45d可见结核菌生长;MDR-TB培养30d终止期茜草素(80μg/ml)单用组可见菌落生长;茜草素(80μg/ml)联合组未见菌落生长;茜草素(80μg/ml)联合组恢复培养45d仍未见菌落生长。结论茜草素在体外对结核分枝杆菌标准菌株(H37RV)表现为抑杀作用,联合抗结核西药对MDR-TB表现为协同作用,可作为治疗耐药株感染的候选药物进一步研究。OBJECTIVE To evaluate the in vitro antimicrobial activity of alizarin against multidrug-resistant Mycobacterium tuberculosis.METHODS The standard strains of Mycobacterium tuberculosis（H37RV）and multidrugresistant tuberculosis（MDR-TB）strains were inoculated in different concentrations of single alizarin,alizarin combined with anti-tuberculosis western medicine,and blank control culture media of M.tuberculosis,the growth situation of M.tuberculosis was observed,and the in vitro antibacterial effect of the alizarin was evaluated.RESULTS The bacterial colonies grew normally in the model group but did not grow in the single alizarin group（80μg/ml）at termination of culture of M.tuberculosis（H37RV）or the rifampicin group（5mg/ml）.The growth of M.tuberculosis was found in the single use groups when the media resumed to be cultured for 45 days.The bacterial colonies grew in the single alizarin group（80μg/ml）at termination of culture of MDR-TB for 30days;the growth of bacterial colonies was not found in the alizarin（80μg/ml）combined group;while the bacterial growth was still not found in the alizarin（80μg/ml）combined group when the media resumed to be cultured for 45 days.CONCLUSION The alizarin shows in vitro killing and suppressing effect on M.tuberculosis（H37RV）,the alizarin combined with anti-tuberculosis western medicine shows synergistic effect;it can be used as an alternative drug for treatment of drug-resistant strains infections,which needs to be further studied.
