摘 要:为了研究极低密度脂蛋白受体(VLDLR)和溶酶体组织蛋白酶(CTSD)的单核苷酸多态性(SNPs),寻找与灵昆鸡产蛋性状相关的候选分子标记,以276只灵昆鸡母鸡为研究对象,采用直接测序法,检测VLDLR和CTSD基因在灵昆鸡中的SNPs,并分析各多态位点不同基因型与11个产蛋性状的相关性。结果表明:VLDLR基因存在1个突变位点C13444T,该位点不同基因型与哈氏单位、蛋黄重及蛋黄比例呈显著相关(P<0.05),在哈氏单位性状中,ZTW型为优势基因型,在蛋黄重及蛋黄比例性状中,ZCW型为优势基因型;CTSD基因有5个突变位点,其中,T2614C对蛋白高度和哈氏单位有显著影响(P<0.05),C9445T、C9475T对500日龄产蛋量和蛋重显著相关(P<0.05),C9457T对蛋重有显著影响(P<0.05),T11028C位点对蛋白高度有显著影响(P<0.05)。C13444T、C9457T、T11028C位点偏离Hardy-Weinberg平衡状态(P<0.05),其他9个位点均处Hardy-Weinberg平衡状态(P>0.05)。结果提示:VLDLR和CTSD基因多态性可能与产蛋性状有关,可尝试作为灵昆鸡产蛋性状分子标记的候选基因,为以后的选育工作提供理论依据。In order to study the single nucleotide polymorphisms(SNPs) of very low density lipoprotein receptor(VLDLR) and cathepsin D(CTSD), and find related candidate molecular markers of egg production traits inLingkun chicken, SNPs of VLDLR and CTSD genes of 276 hens were screened by direct DNA sequencing andits correlation with egg production traits was analyzed to detect the molecular marker of the traits. The resultsshowed that there was a SNP C13444 T in VLDLR gene. The mutation site of C13444 T was significantlycorrelated with Haugh unit(P0.05) with ZTW being the advantage genotype; and it also was significantlycorrelated with yolk weight and yolk proportion(P0.05) with ZCW being the advantage genotype. There were 5SNPs(T2614C, C9445 T, C9457 T, C9475 T, T11028C) in the exon 3, 7, 9 of CTSD gene. The mutation sites ofT2614 C was significantly correlated with Haugh unit and albumen height(P0.05); the mutation sites ofC9445 T, C9475 T were significantly correlated with 500- day- old egg production and egg weight, and themutation site of C9457 T was significantly correlated with egg weight; the mutation site of T11028 C wassignificantly correlated with albumen height. The 3 SNPs(C13444T, C9457 T, T11028C) were not in the Hardy-Weinberg equilibrium, the others were in Hardy- Weinberg equilibrium. The results indicated that eggproduction traits were correlated with VLDLR and CTSD genes which could be the candidate molecularmarkers, and provided the basic data for the breeding of Lingkun chicken.
