益气除痰方调控JNK/SAPK信号通路干预A549细胞的试验研究

摘　　要：目的:研究益气除痰方含药血清对人肺腺癌A549细胞增殖、凋亡及对JNK、p-JNK等表达的影响。方法:采用MTT法检测益气除痰方含药血清对A549细胞增殖的影响,流式细胞术(Annexin V-FITC/PI双染色法)测定细胞凋亡率,Western blotting检测JNK和p-JNK的蛋白表达。结果:MTT结果显示益气除痰方含药血清可抑制A549细胞增殖,且具有一定的时间-浓度依赖性;流式细胞术结果显示益气除痰方含药血清可诱导A549细胞凋亡;Western blotting结果显示益气除痰方含药血清干预A549细胞后可减少p-JNK的蛋白表达,但对JNK蛋白表达无明显影响。结论:益气除痰方含药血清可抑制A549细胞增殖,诱导A549细胞凋亡,其机制可能与抑制JNK/SAPK信号通路有关。Objective： To study the effect of cell proliferation,apoptosis,JNK,p-JNK of human lung carcinoma cell line A549 cells intervened by the drug serum of Formula of Invigorating Qi and Expelling Phlegm. Methods： The effect of A549 cell proliferation intervened by the drug serum of Formula of Invigorating Qi and Expelling Phlegm were detected by MTT assay,apoptosis rates were measured by flow cytometry（ Annexin V-FITC / PI double staining）,JNK and p-JNK protein expressions were detected by western blotting. Results： The result of MTT assay showed that the drug serum of Formula of Invigorating Qi and Expelling Phlegm can suppress proliferation of A549 cell,the growth inhibition effects of each concentration were enhancing with the extension of time; the result of flow cytometry showed that the drug serum could induce cell apoptosis of A549cell; the result of western blotting shows that the drug serum can reduce the expressions of p-JNK protein of A549 cell,but the expressions of JNK protein of A549 cell were of no statistical significance. Conclusion： The drug serum of Formula of Invigorating Qi and Expelling Phlegm can suppress proliferation and induce cell apoptosis of A549 cell. Its mechanism is probably related to inhibit JNK / SAPK pathway.
