摘 要:目的体外建立胰岛素抵抗Hep G2细胞模型,可用于中药有效成分的胰岛素抵抗活性筛选研究。方法用不同浓度的胰岛素对Hep G2细胞进行诱导后进行不同作用时间的筛选,通过葡萄糖氧化酶法对Hep G2细胞的葡萄糖消耗量及CCK-8法对细胞活性评价,明确最优的Hep G2细胞胰岛素抵抗模型建立的浓度和作用时间。结果用10^(-6) mol/L的胰岛素诱导处理36 h是Hep G2细胞产生胰岛素抵抗的最适条件。结论高胰岛素培养法可以建立Hep G2细胞胰岛素抵抗模型,具有建模周期短、价格低、易于重复、可控性强的优点,可用于抗胰岛素抵抗中药成分的筛选研究。Objective To establish human hepatoma carcinoma cell (HepG2 cell) model of insulin resistance in vitro and to initially select the effective ingredients in traditional Chinese medicine for improving insulin resistance. Methods HepG2 cells were induced by insulin of different concentrations at different time. Through evaluating glucose consumption with glucose oxidase method and the cell activity with cck-8 method, the best concentration and induction time of insulin that can induce the insulin resistance cell model were ascertained. Conclusion High concentration of insulin could successfully induce insulin resistance in HepG2 cells with the advantages of time saving, good repeatability and ease of operation. This model is also applicable to screening of traditional Chinese medicine that improves insulin sensitivity.
