摘 要:目的研究大孔树脂分离、纯化葛根中总异黄酮的工艺条件。方法以3’-羟基葛根素、3’-甲氧基葛根素、葛根素、大豆苷元-8-C-芹菜糖-(1→6)葡萄糖苷、大豆苷、大豆苷元6种成分的吸附量和解吸率为评价指标,考察9种不同类型树脂。并对优选出的树脂纯化葛根总异黄酮的工艺条件及参数进行考察。结果 HPD600大孔树脂吸附、解吸效果最好,上柱药液浓度为75 mg/ml,吸附上样流速1BV/h,径高比1:10,除杂溶剂为水,除杂体积5 BV,除杂流速1 BV/h,洗脱溶剂为50%乙醇,洗脱体积6BV,洗脱流速2 BV/h。结论葛根中6种异黄酮成分精制后含量保留均在80%以上,总异黄酮纯度在60%以上,表明优选的AB-8大孔树脂纯化葛根总异黄酮工艺是可行的。Objective To investigate the process of isolation and purification of total isoflavonoids from Puerariae lobatae by macroporous adsorption resin. Methods The adsorption amount and desorption ratio of the six kinds of isoflavonoids were chosen as indexes for refining to evaluated nine kinds of macroporous adsorption resins.Also the process conditions and parameters of total isoflavonoids from Puerariae lobatae by optimized macroporous adsorption resin were researched. Results The consequence is type of HPD600 had the best dynamic adsorptivity and separability to the total isoflavonoids from Puerariae lobatae when the content in the aqueous extract was 75 mg per 1 ml equivalent to raw material, the ratio of diameter and height was 1:10, velocity of adsorption was 1 BV/h, solvent of removing impurities was water, volume of removing impurities was 5 BV, velocity of removing impurities was 1 BV/h, solvent of elution was 50% of ethanol, volume of elution was 6 BV, velocity of elution was 2 BV/h. Conclusion The content of six kind isoflavonoids in Puerariae lobatae were retained more than 80%, besides the purity of total isoflavonoids was over 60% after isolation and purification by macroporous adsorption resin. It is concluded that the optimized process is feasible.
