摘 要:目的探讨壮药绞股蓝Gynostemma pentaphyllum正丁醇提取物对快速老化SAMP8小鼠淀粉样蛋白(Aβ蛋白)的影响。方法 50只快速老化SAMP8小鼠随机分为模型组、石杉碱甲组[0.15 mg/(kg·d)]和绞股蓝正丁醇提取物高、中、低剂量组[500、250、125 mg/(kg·d)],10只正常老化SAMR1小鼠为正常对照组。灌胃给药8周后,取小鼠血清和脑匀浆,以ELISA法检测其Aβ蛋白的水平,RT-PCR和免疫组织化学法检测App基因和APP蛋白的表达。结果与模型组比较,绞股蓝正丁醇提取物给药组小鼠脑组织中的APP基因表达和蛋白表达明显下调,血清和脑组织中的Aβ蛋白水平显著降低。结论绞股蓝正丁醇提取物可通过抑制APP蛋白的生成,使Aβ蛋白的表达减少,从而起到防治阿尔茨海默症(AD)的作用。AIM To explore the effect of n-butanol extract from Gynostemma pentaphyllum,a native herb of the Zhuang nationality,on the expression of Aβ protein in SAMP8 senescence-accelerated mice. METHODS Fifty SAMP8 senescence-accelerated mice were randomly divided into five groups,model,positive drug [Huperzine-A,0. 15 mg /( kg·d) ],high-,medium- and low-dose of n-butanol extract from Gynostemma pentaphyllum groups [500,250,125 mg /( kg·d) ]. Furthermore,ten SAMR1 normal aging mice were used as normal control.After 8-week intragastric administration,the serum and brain homogenate of mice were collected to detect the Aβprotein level with enzyme linked immunosorbent assay( ELISA),App gene expression by real-time polymerase chain reaction( RT-PCR),and APP protein expression using immunohistochemistry. RESULTS Compared with the model group,the lower expressions of App gene and APP protein in brain tissue of mice could be observed in each group treated with n-butanol extract from Gynostemma pentaphyllum. This result was accompanied by significantly reduced Aβ protein levels in serum and brain tissue. CONCLUSION The n-butanol extract from Gynostemma pentaphyllum can restrain APP protein formation,and reduce Aβ protein expression,which plays a role in the prevention and treatment of Alzheimer's disease.
