摘 要:目的:考察茵黄合剂提取物(YHA)对肝内胆汁淤积模型大鼠胆汁分泌和肝脏功能的改善作用。方法:将SD大鼠随机分为空白组、模型组、阳性组(熊去氧胆酸,45 mg/kg)和YHA高、中、低剂量组[37.8、18.9、9.45 g(生药)/kg],每组10只。各给药组大鼠ig相应药物,空白组和模型组大鼠ig等容积生理盐水,每天1次,连续5 d。给药3 d后,除空白组外,其余各组大鼠均单次ig给予α-异硫氰酸萘酯诱导肝内胆汁淤积模型。末次给药1 h后,检测大鼠2 h胆汁流量和血清中总胆红素(TBIL)、总胆汁酸(TBA)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)及碱性磷酸酶(ALP)水平;观察大鼠肝脏病理变化。结果:与空白组比较,模型组大鼠2 h胆汁流量明显减少,血清中TBIL、TBA、ALT、AST及ALP水平显著升高(P<0.01);肝组织病理性损伤明显。与模型组比较,除阳性组和YHA中、低剂量组大鼠血清中ALP水平以及YHA低剂量组大鼠血清中TBIL水平降低不显著外,其余各组大鼠各指标均明显改善(P<0.05或P<0.01);除YHA低剂量组外,其余各给药组大鼠病理损伤得到明显改善。结论:YHA对α-异硫氰酸萘酯致肝内胆汁淤积模型大鼠的胆汁分泌和肝脏功能具有一定的改善作用。OBJECTIVE: To investigate the improvement effects of Yinhuang mixture extract (YHA) on biliation and liver function of intrahepatic cholestasis model rats. METHODS: SD rats were randomly divided into blank group, model group, positive group (ursodesoxyeholic acid, 45 mg/kg) and YHA high-dose, medium-dose and low-dose groups [37.8, 18.9, 9.45 g(crude drug)/kg], with 10 rats in each group. Treatment groups were given relevant medicine intragastrically, and blank group and model group were given constant volume of normal saline intragastrically, once a day, for consecutive 5 d. 3 d after administration, except for blank group, those groups were given a-naphthyl isothiocyanates intragastrically to induce intrahepatic cholestasis model. 1 h after last administration, 2 h volume of bile, serum levels of TBIL, TBA, ALT, AST and ALP were detected; pathological changes of liver were observed. RESULTS: Compared with blank group, the 2 h volume of bile decreased significantly in model group, and serum levels of TBIL, TBA, ALT, AST and ALP increased significantly (P〈0.01). The pathological injury was found in liver tissue. Compared with model group, except for serum level of ALP in positive group, YHA low-dose and medium-dose groups and serum level of TBIL in YHA low-dose group, another indexes of other treatment groups were improved significantly (P〈0.05 or P〈0.01). Except for YHA low-dose group, pathological injure of other treatment groups were improved significantly. CONCLUSIONS: YHA can improve the biliation and liver function of rats with intrahepatic cholestasis induced by α-naphthyl isothiocyanates.
