摘  要:目的:通过研究跑台运动对雌激素剥夺大鼠的抑郁样行为及前额叶皮质nNOS表达的变化,以期探讨跑台运动治疗抑郁症的可能机制。方法:30只成年SD雌性大鼠随机分为假手术组(sham group,n=10)、卵巢切除组(OVX group,n=10)及去卵巢运动组(OVX+Ex group,n=10)3组。跑台运动8周后通过强迫游泳实验观察运动对雌激素剥夺大鼠抑郁样行为的影响;然后用免疫组织化学结合图像半定量方法测量和分析前额叶皮质nNOS神经元的表达。结果:与假手术组大鼠比较,强迫游泳实验中OVX组大鼠游泳不动时间显著延长(P<0.01),攀爬时间及攀爬次数均显著缩短(P<0.01);前额叶皮质nNOS表达显著上调(P<0.01);8周跑台运动显著改善了去卵巢大鼠的抑郁样行为,前额叶皮质nNOS表达显著下降(P<0.01)。结论:中等强度跑台运动显著改善了去卵巢大鼠的抑郁样行为,可能与此运动削弱去卵巢大鼠前额叶皮质nNOS的表达,降低NO水平拮抗HPA轴功能亢进有关。Objetive:To observe the effects of treadmill exercise on the depressive behavior and prefrontal cortex nNOS expression in estrogen-deprived female rats and explore the possible mechanism.Methods:30adult SD female rats were randomly assigned to 3groups:SHAM(sham operation group,n=10);OVX(ovariectomy group,n=10)and OVX+Ex(ovariectomy rats treated with exercise group).The behavior changes were observed by forced swimming test(FST)after 8week treadmill exercise.Subsequently,immunohistochemical staining was used to detect nNOS expression in prefrontal cortex.Results:Compared with the sham group rats,immobility time prolonged(P〈0.01),climbing time(P〈0.01)and numbers(P〈0.01)decreased in FST;prefrontal cortex nNOS expression significantly enhanced(P〈0.01);8week treadmill exercise ameliorated depression-like behavior and reduced prefrontal cortex nNOS expression in OVX+Ex group rats when compared with the OVX group.Conclusion:Our results suggested that treadmill exercise amelioration of depression-like behavior in OVX rats is partly due to reduced prefrontal cortex nNOS expression,decreased NO level and regulate HPA axis dysfunction.
