摘 要:目的:研究白术对脾虚动物利水作用,探讨其可能作用机制。方法:采用优化后饮食不节结合劳倦过度法,造成动物脾虚模型。腹腔水负荷生理盐水建立脾虚水湿内停模型。肛温、体重、表观指标为脾虚模型大鼠的造模期评价指标。尿中D-木糖排出量、粪便含水率、血清总蛋白(TP)、白蛋白(ALB)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、水通道蛋白1、胃泌素(Gas)及肾髓水通道蛋白1(AQP1)等为研究指标,探讨白术调节脾虚大鼠胃肠功能的有效剂量及相关机制。结果:造模期,大鼠体重下降,肛温无变化,表明此方法造成动物脾虚。给药后,白术(3.6 g·kg-1)能略增模型大鼠尿量和尿中D-木糖排出量,明显升高粪便含水量、TP、ALB、HDL-C,但降低TC和LDL-C含量,略降低水通道蛋白1水平。腹腔水负荷试验,模型组动物,体重下降率显著降低,给予白术水煎液后则下降率显著升高,尿量略升高。结论:中剂量组白术水煎液有运化水湿作用,可有效改善动物模型脾虚状态,提高其相应消化吸收功能。初步认为白术排水途径可能由粪便排出。对大鼠进行腹腔水负荷后,说明白术有改善机体水液代谢能力,能有效改善TP、ALB、HDL-C、TC、LDL-C等指标。白术对水的调节作用不通过AQP1实现,且白术高剂量效果不及中剂量。Objective:To observe the effect of Atractylodes macrocephala Koidz. for alleviating the water retention in the rat model with the spleen deficiency. Methods : The optimized methods of improper diet and overstrain were used to replicate spleen-deficiency model. The rats with spleen deficiency were further administered intraperitoneal ( i.p. )water to prepare the cohesion model of dampness. The changes of the rectal temperature, weight and the apparent situation of the dampness cohesion model was used to evaluate the effective substances of Atractylodes macrocephala Koidz.. The concentration of D-xylose in urine, the water ratio of faeces, the TP, ALB, TC, HDL-C, LDL-C and GAS in serum, AQP1 in the renal medullary part were determined. The loss ratios of body weight and the volume of urine were used as main index in the experiment of cohesion model of dampness to elucidate the effective fractions of Atraetylodes macroeephala Koidz.. Results : During the period of model preparation, the weight of rats was decreased and no change was observed in rectal temperature, indicating that spleen deficiency had formed. The administration with the water decoction ofAtraetylodes macrocephala Koidz. at 3.6 g · kg-1 dose could improve the volume of urine slightly, increase the concentration of the D-xylose in urine, the water ratio of faeces, the content of TP, ALB and HDL-C in serum. Then the decoction could reduce the level of AQP1 slightly and could reduce the concentration of TC and LDL-C significantly. In the test of the water load in the abdominal cavity, the loss ratio of body weight of the model group decreased obviously. After administered with Atractylodes maerocephala Koidz. water decoction, the loss ratio of body weight increased significantly. But the volume of urine increased slightly, no obvious changes. Conclusions : Middle dose of Atractylodes macrocephala Koidz. water decoction has the effect on transmitting and distributing the dampness in spleen deficiency animals.
