摘 要:目的:探讨野生与栽培楤木芽水提物、总皂苷及其化学成分楤木皂苷A对脂肪变性肝细胞L-02脂代谢的干预作用。方法:采用含医用脂肪乳的培养液诱导建立肝细胞脂肪变性模型,分别给予高浓度组(100μg/m L)、中浓度组(50μg/m L)和低浓度组(10μg/m L)野生与栽培楤木芽水提物、野生与栽培楤木总皂苷和楤木皂苷A进行培养,24 h后测定肝细胞内甘油三酯(TG)、胆固醇(TC)的含量。结果:该研究细胞造模成功。与对照组比较,模型组TC和TG含量均显著升高(P〈0.05)。与模型组比较,野生楤木芽总皂苷、栽培楤木芽总皂苷和楤木皂苷A均可降低TC的含量(P〈0.05),其中栽培楤木芽总皂苷低浓度组降低TC效果优于其他组(P〈0.05),且在10~100μg/m L范围内呈现量效关系;野生楤木芽总皂苷(低、中浓度组)、野生楤木芽水提物(中浓度组)和楤木皂苷A(低、中、高浓度组)可降低细胞内TG的含量(P〈0.05)。结论:楤木芽总皂苷和楤木皂苷A对体外肝细胞脂肪变性模型可产生积极的脂代谢影响,具有一定的潜在开发价值。Objective:To observe the intervention effects of aqueous extract, total saponins and the chemical constituent of araloside A from the bud of cuhivated and wild Aralia chinensis on the lipid metabolism of steatosis hepatocyte L-02. Methods : The steatosis hepatocyte L-02 model was established by induction of the medium containing fat emulsion. The hepatocytes in the treatment group were treated with the water extraction, the total saponins and the compound of araloside A from the bud of cultivated and wild Aralia chinensis which divided into high concentration group ( 100 Ix g/mL ), medium concentration group ( 50 p~ g/mL ) and low concentration group ( 10 Ix g/mL ), respectively. The triglyceride ( TG ) and total cholesterol ( TC ) contents were determined after 24-hour incubation. Resuhs : This study cells building successful compared with control group, model group TC and TG content were significantly higher ( P〈0.05 ). Compared with model group, the total saponins from the bud of wild and cultiviated Aralia chinensis, araloside A can reduce the content of TC ( P〈0.05 ), of which low concentration of the total saponins from the bud of cultiviated Aralia chinensis group lower TC is better than other groups ( P〈0.05 ), and within 10 - 100 Ix g/mL present concentration-response relationship; total saponins from the bud of wild Aralia chinensis ( low and medium concentration groups ), water extract from the bud of wild Aralia chinensis ( medium concentration group ) and araloside A ( high, medium and low concentration groups ) can reduce the content of intracellular TG ( P〈0.05 ). Conclusion : The total saponins from the bud of Aralia chinensis and araloside A have positive effect on lipid metabolism of steatosis hepatocyte L-02 in vitro, of certain potential development value.
