摘  要:目的研究血管活性因子结缔组织生长因子(Connective tissue growth factor,CTGF)对大鼠血管外膜成纤维细胞(Adventitial fibroblasts,AFs)增殖作用的影响和细胞内信号转导机制。方法离体培养AFs并作鉴定,采用CCK-8技术确定CTGF促AFs增殖作用的效应浓度,在CTGF效应浓度下加入不同的细胞内信号转导阻断剂,观察哪种信号通路参与CTGF促AFs的增殖效应。结果采用组织贴块法可以培养出AFs,经免疫荧光检测培养的细胞为AFs,CTGF以浓度依赖方式促进AFs增殖,CTGF促AFs增殖的效应能被Rho激酶阻断剂Y-27632、钙调神经磷酸酶(Calcineurin,Ca N)阻断剂环孢菌素A(Cyclosporine A,CSA)和钙通道阻断剂尼卡地平所阻断。结论贴块法能成功培养出AFs;CTGF是可以促进AFs增殖的趋化因子,其促进增殖效应可能通过Ca N、Rho以及Ca2+通路来介导。Objective To study the effect of vasoactive connective tissue growth factor (CTGF) on adventitial fibroblasts (AFs) proliferation and explore the mechanism of signal transduction. Methods AFs in vitro were cultured and cellular components were identified;CCK-8 technology was used to observe the effect of CTGF on proliferation of AFs;different intracellular signal transduction inhibitors were added to observe which signaling pathways involved in the proliferation effect of CTGF on AFs. Results The cells cultured by explant patch method were identified as AFs by immunofluorescence assay. CTGF promoted AFs proliferation in a concentration dependent manner. CTGF's promoting effect on the proliferation of AFS could be blocked by Rho kinase blocking agent Y-27632, calcineurin bloc- king agent cyclosporine A (CSA) and calcium channel blocker nicardipine. Conclusion AFs can be successfully cultured by the explant patch method, and CTGF can promote the proliferation of AFs, which may promote the proliferation effect through CaN, Rho and Ca2+ pathway.
