摘 要:探讨了17β-雌二醇(17β-E2)对体外培养大鼠海马神经元细胞内钙离子水平([Ca^(2+)]i)的快速影响及其可能的机制.大鼠海马神经元体外培养8~10 d,用Fluo 4-AM钙离子荧光探针负载胞内Ca^(2+),用激光共聚焦显微镜动态检测了17β-E2对谷氨酸诱导的神经元内Ca^(2+)荧光强度的影响.结果显示:17β-E2(10 nmol·L^(-1))处理30 min,对正常状态下神经元的[Ca^(2+)]i无显著影响,但可快速促进谷氨酸诱导的[Ca^(2+)]i增加(与对照组相比,P<0.001),雌激素受体阻断剂ICI182780(10μmol·L^(-1))预处理可阻断17β-E2的这一促进作用;胞外信号调节激酶(ERKs)抑制剂U0126(10μmol·L^(-1))单独处理神经元,可极显著抑制谷氨酸诱导的[Ca^(2+)]i增加(P<0.001),但这一作用可以被17β-E2部分逆转.研究结果提示,ERKs信号通路可能参与谷氨酸诱导的神经元[Ca^(2+)]i增加,17β-E2可能经雌激素受体介导的ERKs信号通路快速促进谷氨酸诱导的神经元[Ca^(2+)]i增加.It was investigated the effects of 17β-estradiol ( 17β-E2) on calcium level in hippocampal neurons of rats. After in vitro cultured for 8 - 10 days, the hippocampal neurons were marked by Fluo 4-AM, an intra- cellular Ca2+ fluorescent probe. Laser confocal scanning microscopy was used to detect the dynamic change of glutamate-induced intrareellular calcium concentration ([ Ca2+ ]i}. These results showed that treatment of 17β-E2 (10 nmol· L-1 )for 30 min had no remarkable effect on [ Ca2+ ]i of neurons in the normal condition, but rapidly enhanced the increase in the [ Ca2 ~ ] i induced by glutamate ( P 〈 0. 001, compared with control }. The pretreatment of estrogen receptor antagonist, ICI182780 ( 10μmol·L-1 ) blocked this effect of 17β-E2. ERKs inhibitor, U0126 (10μmol·L-1) significantly suppressed glutamate-induced increase of [ Ca2+ ]i (P 〈 0. 001), suggesting ERKs signal participates in glutamate-induced calcium influx; however, this effect was reversed by pretreatment of 17β--E2. These results suggested that 17β--E2 could rapidly enhanced the increase in [ Ca2+]i induced by glutamate in hippocampal neurons in vitro. These effects might be mediated by ERKs signaling pathway via estrogen receptor.
