摘 要:目的:探讨红景天对大鼠心肌梗死后血管新生过程中肾上腺髓质素(adrenomedullin,ADM)表达的调控作用。方法:将8周龄雄性Wistar大鼠分为4组,正常对照组、假手术组、心肌梗死造模后生理盐水对照组(生理盐水组)及心肌梗死造模后红景天干预组(红景天组)。饲养1周后处死动物,摘取心脏制作石蜡切片,采用CD31免疫组化染色进行微血管标记染色;处死动物前留取血浆,采用ELISA法测定血浆ADM浓度;Western blot检测不同组心肌组织ADM的蛋白表达水平。结果:与正常对照组、假手术组及生理盐水组相比,红景天组大鼠心肌梗死边缘区微血管密度明显升高;红景天组(1.06±0.01)及生理盐水组(0.52±0.09)的缺血心肌组织ADM蛋白表达水平较正常对照组及假手术组均明显升高(P<0.05),红景天组较生理盐水组ADM蛋白表达水平进一步升高(P<0.05);红景天组[(32.28±5.13)ng/L]和生理盐水组[(28.55±3.49)ng/L]血浆ADM水平较正常对照组及假手术组均明显升高(P<0.05),但红景天组和生理盐水组之间无明显差异。结论:心肌梗死后大鼠血浆ADM水平变化与心肌内不同步,红景天对ADM表达的上调作用主要在缺血心肌,具有一定的靶向性。Objective: To explore the regulative effect ot rhodlota on me expresstorl ut adrenomedullin(ADM) during angiogenesis of rats after myocardial infarction. Methyls.. The eight- week-old Wistar rats were divided into the normal control group, the sham operated group, the saline group and the rhodiola group. They were sacrificed after one week feeding. Their hearts were resected and embedded in paraffin to make sections. Microvessels were marked with CD31 immunohistochemical staining. Plasma concentration of ADM was tested by ELISA before sacrifice. The protein expression levels of ADM in myocardial tissue among different groups were observed and compared by western blot. Results:The rhodiola enhanced the rat's microvascular density in infarcted border zones after treatment, showing significant difference as compared with saline group, sham-operated group and normal control group. The protein expressions of ADM in ischemic myocardial tissue after rhodiola (1.06 ± 0. 10) and saline treatment (0.52 ± 0.09) were significantly higher than the other two groups (P^0. 05), while rhodiola further up-regulated the ADM expression than saline (P(0. 05). Plasma concentrations of ADM in rhodiola group [(32. 28 ± 5. 13) ng/L and saline group [(28. 55 ± 3.49) ng/L] were higherthan those in the other two groups (P〈0. 05), while no significant difference was observed between rhodiola group and saline group. Conclusion:The rhodiola can up-regulate ADM expression in ischemic myocardial tissue. The change in ADM plasma concentration doesn't synchronize with the altered ADM expression in myocardial tissue after myocardial infarction. The up-regulatory effect of rhodiola on ADM expression is mainly focused on the ischemic myocardial, showing a targeting characteristic.
