前痛宁方对慢性非细菌性前列腺炎模型大鼠细胞因子TNF-α、IL-8、IL-10及β-EP的影响

摘　　要：目的:观察前痛宁方对慢性非细菌性前列腺炎(CAP)模型大鼠前列腺组织细胞因子TNF-α、IL-8、IL-10及β-EP的影响。方法:将40只SD雄性大鼠随机分成空白组、模型组、盐酸坦洛新组和前痛宁方组。对除空白组外的其余3组大鼠行去势手术后于背部皮下注射苯甲酸雌二醇法制作CAP模型,各组分别给予相对应的药物干预。检测比较各组前列腺组织TNF-α、IL-8、IL-10及β-EP的含量。结果:模型组大鼠前列腺组织TNF-α、IL-8含量较空白组明显升高(P<0.01),β-EP含量较空白组明显降低(P<0.01);盐酸坦洛新组β-EP含量较模型组明显升高(P<0.01);前痛宁方组TNF-α、IL-8含量均明显低于模型组和盐酸坦洛新组(P<0.01),IL-10、β-EP含量明显高于模型组和盐酸坦洛新组(P<0.01,P<0.05)。结论:多种细胞因子参与了CAP的发病过程;中药前痛宁方可能通过降低前列腺组织内促炎性因子TNF-α、IL-8的水平,升高抗炎性因子IL-10及β-EP的水平,从而减轻前列腺组织局部的炎症反应,缓解疼痛,促进前列腺组织的修复。Objective： To study the effects of Qiantongning formula on tumor necrosis factor-α（ TNF-α）,interleukin-8（ IL-8）,interleukin-10（ IL-10）,and β-endorphin（ β-EP） in a rat model of chronic aseptic prostatitis（ CAP）. Methods： Forty Sprague-Dawley rats were randomly divided into blank control group,model group,tamsulosin hydrochloride group,and Qiantongning formula group. All the rats except those in blank control group were subcutaneously injected with estradiol benzoate in the back following castration to establish the CAP model. Then these groups were given respective treatments,and the levels of TNF-α,IL-8,IL-10,and β-EP in prostate tissue were measured and compared. Results： Compared with the blank control group,the model group had significantly increased levels of TNF-α and IL-8 in prostate tissue（ P〈0. 01）,but a significantly reduced β-EP level（ P〈0. 01）. The tamsulosin hydrochloride group had a significantly higher β-EP level than the model group（ P〈0. 01）. Compared with the model group and tamsulosin hydrochloride group,the Qiantongning formula group had significantly lower TNF-α and IL-8 levels（ P〈0. 01）,but significantly higher IL-10 and β-EP levels（ P〈0. 01,P〈0. 05）. Conclusion： Multiple cytokines are involved in the pathogenesis of CAP. Qiantongning formula possibly reduces the proinflammatory cytokines TNF-α and IL-8 and increases the anti-inflammatory cytokines IL-10 and β-EP in prostate tissue to alleviate focal inflammations in the prostate,relieve pains,and promote prostate tissue repair.
