摘  要:通过高温加热法,以天然凹土、硅酸钠、氯化铝为原料,十六烷基三甲基溴化铵(CTAB)为模板剂,在碱性条件下制备凹土介孔材料,并将其用于固载猪胰脂肪酶(PPL)。通过扫描电子显微镜(SEM)、N_2吸附脱附分析仪(BET)、傅里叶红外光谱(FT-IR)等手段表征制备的介孔材料。结果表明:合成的样品具有典型的介孔结构,凹土表面呈现较大孔径。凹土介孔材料的比表面积为102.27 m^2/g。实验表明脂肪酶固定化的最适温度为25℃,最适pH为6.0,固载4 h后固定化脂肪酶的热稳定性、pH稳定性均优于游离酶。Attapulgite mesoporous materials were synthesized via high temperature method, with natural attapulgite,sodium silicate and aluminum chloride as raw materials and cetyl trimethyl ammonium bromide (CTAB)as a template agent under alkaline condition.Then the synthesized material was used to immobilizae porcine pancreatic lipase(PPL).Immobilization samples were characterized by scanning electron microscope (SEM),Brunauer?Emmet?Teller (BET) adsorption and desonsion isotherm and Fourier transform infrared spectroscopy(FT?IR),et al.Results show that the synthesized sample had typical mesoporous structure and larger aperture on its surface. Attapulgite mesoporous materials had a surface area of 102. 27 m^2/g. The optimum temperature for lipase immobilization was 25℃,and the optimum pH was 6. The thermal stability and pH stability of immobilized lipase were better than free enzyme after loading for 4 h.
