摘 要:目的:研究钙调蛋白激酶Ⅱ( CaMK Ⅱ)抑制剂( KN-93)对异丙肾上腺素( ISO)作用下心肌细胞电生理特性的影响。方法酶解法急性分离小鼠心室肌细胞,将心肌细胞分为4组:正常对照组、异丙肾上腺素干预组( ISO组)、异丙肾上腺素+KN-92干预组( ISO+KN-92组)、异丙肾上腺素+KN-93干预组( ISO+KN-93组);应用全细胞膜片钳技术研究ISO及KN-93对心肌细胞L-型钙电流( ICaL )的影响;应用激光扫描共聚焦显微技术记录ISO及KN-93作用下心肌细胞钙瞬变;western bolt检测相关蛋白的表达水平。结果与对照组相比,ISO使ICaL峰值电流密度增大[ISO组:(-4.25±0.99) pA/pF,对照组:(-2.66±1.31) pA/pF,P=0.01]:L型钙通道激活加速[ISO组半数激活电压:(-0.43±0.66) mV,对照组半数激活电压:(3.94±1.58) mV,P<0.01]、失活加速[ISO组半数失活电压:(-10.91±1.11) mV,对照组半数失活电压:(-8.03±0.58) mV, P<0.01],并缩短失活后恢复时间[ ISO组恢复时间常数:(82.74±2.21) ms,对照组恢复时间常数:(119.32±4.24) ms,P<0.01];ISO使钙瞬变幅值增大(0.5 HZ:ISO组2.08±0.72,对照组1.05±0.43,P<0.01;1.0 HZ:ISO组1.95±0.75,对照组1.07±0.41,P<0.01),易诱发钙紊乱(P<0.01)。 KN-93减弱ISO引起的钙稳态变化,使ICaL峰值电流密度减小(P<0.05),激活减慢(P<0.01),延长失活后恢复时间(P<0.01);并使钙瞬变幅值减小(0.5 HZ:P<0.05,1.0 HZ:P<0.05),减少钙紊乱。结论 KN-93能调节ISO引起的心肌细胞电生理特性的改变,在预防心律失常中发挥作用。Objective To study the effect of isoproterenol ( ISO ) and calmodulin kinase Ⅱ( CaMKⅡ)inhibitor(KN-93)on the electrophysiological characteristics of myocardial cells .Methods Mouse ventricu-lar myocytes were acutely isolated by enzymatic method .Cardiomyocytes were divided into four groups:control group,ISO group,ISO+KN-92 group,ISO+KN-93 group.The whole-cell patch clamp was used to study the effect of ISO and KN-93 on L-type calcium current ( ICaL ) in isolated ventricular myocytes .The technology of Laser scanning confocal microscopy was used to record cardiomyocytes calcium transients after ISO and KN -93 were used.Western blot was used to detect the expression levels of related proteins .Results ISO significantly in-creased current density of ICaL[control:(-2.66±1.31) pA/pF vs.ISO:(-4.25±0.99) pA/pF,P〈0.01],and decreased the half activation voltage [control:(3.94±1.58) mV vs.ISO:(-0.43±0.66) mV,P〈0.01],half inactivation voltage[control:(-8.03±0.58) mV vs.ISO:(-10.91±1.11) mV,P〈0.01]and the recovery time constant[control:(119.32±4.24) ms vs.ISO:(82.74±2.21) ms,P〈0.01].In the presence of CaMK Ⅱ in-hibitor,KN-93 decreased the increased current density of ICaL(P〈0.05)and prolonged the shortened recovery time constant caused by ISO(P〈0.01).In addition,KN-93 alone can change the activation,inactivation and re-covery kinetics of ICaL .Moreover ISO significantly increased the Ca 2+transient amplitude during both stimulation frequencies(0.5 HZ:P〈0.01,1.0 HZ:P〈0.01)and was easy to induce calcium disorders .But in the presence of KN-93,these changes were weakened (0.5 HZ:P〈0.05,1.0 HZ:P〈0.05).Conclusion Changes of the myocardial electrophysiological properties caused by ISO can be adjusted by KN -93,thus KN-93 plays a role in the prevention of arrhythmias .
