摘  要:目的研究采用仿生原理制备的胶原基人工骨基质的理化性能。方法用Ⅰ型胶原、饱和二水氯化钙制备胶原基人工骨基质。采用扫描电子显微镜、金相显微镜、傅里叶变换红外光谱(FT-IR)分析、差热-热重(TG-DTG)分析、X射线衍射(XRD)分析、X射线能谱仪(EDS)等分析测试手段对其形貌、组成、结晶度和Ca/P等进行详细的研究。结果采用扫描电子显微镜和金相显微镜观察,胶原基人工骨基质无机钙盐均匀沉积在自组装的胶原模板上,有清晰的孔隙,呈梯度分布,并且具有类似天然骨的内连孔结构;孔隙直径为50~500μm;孔隙率为(68±5)%。FT-IR分析,胶原基人工骨基质与天然骨的红外图谱基本吻合,胶原基人工骨基质的872 cm^(-1)为HPO_4^(2-)的吸收峰。TG-DTG分析证明有羟基磷灰石。XRD、EDS分析,无机相主要为羟基磷灰石,与天然骨的衍射峰位置、数量相似。结晶度和Ca/P分别为49.3%、1.64。结论仿生原理制备的胶原基人工骨基质与人骨基质相似。Objective To investigate the physicochemical properties of the collagen-based artificial bone matrix prepared according to bionic principle. Methods The collagen based artificial bone matrix was synthesized by type Ⅰ collagen and saturated calcium chloride dihydrate. The morphology, composition, crystallinity and Ca/P of the collagen-based artificial bone ma-trix were characterized by scanning electron microscope(SEM), metallographic microscope, Fourier transform infrared spectroscopy(FT-IR), differential thermal gravimetric analysis(TG-DTG), X-ray diffraction(XRD), X-ray energy dispersive analysis(EDS). Results SEM and metallographic microscope analysis result showed that the collagen-based artificial bone matrix 's inorganic calcium salt uniformly deposited on the self-assembled collagen template and the bone porosity of the collagen-based artificial bone matrix was(68 ± 5) %, the gradient distribution of clear pores showed pore diameter of 50 to 500 microns,with inner connection structure similar to natural bones. FT-IR analysis showed that the collagen-based artificial bone matrix had similar infra-red spectrum with natural bone, and the absorption peak at 872 cm^-1 was ascribed to HPO4^2-. TG-DTG result proved the existence of hydroxyapatite. XRD and EDS test results showed that the inorganic phase was mainly hydroxyapatite, the position and number of the diffraction peaks was similar to that of natural bone. Crystallinity and Ca/P were 49.3 %and 1.64. Conclusion The collagen based artificial bone matrix prepared by bionic principle is similar to human bone.
