健脾疏肝和胃方对功能性消化不良大鼠Cajal间质细胞及胃排空的影响

摘　　要：目的:观察健脾疏肝和胃方对功能性消化不良(functional dyspepsia,FD)大鼠胃排空(gastric emptying,GE)、c-Kit阳性Cajal间质细胞(interstitial cells of cajal,ICC)及其表达的影响,探讨其可能的作用机制。方法:将大鼠随机分成对照组、模型组、健脾疏肝和胃方组、多潘立酮组,采用夹尾刺激法建立FD大鼠模型,连续给药14天后,采用半固体营养糊法观察胃排空,胃高倍镜下Cajal间质细胞计数,采用实时荧光定量PCR法检测胃平滑肌组织kit mRNA表达,Western blot法检测胃平滑肌组织kit表达。结果:与对照组相比,模型组胃排空延迟、胃底c-Kit阳性ICC表达显著降低(P均<0.05);与模型组相比,健脾疏肝和胃方组及多潘立酮组胃排空、c-Kit阳性ICC数量及表达显著升高(P均<0.05)。结论:健脾疏肝和胃方组可以明显增加FD大鼠胃底ICC表达,加快胃排空;上调ICC、提高大鼠胃排空率可能是健脾疏肝和胃方治疗功能性消化不良的作用机制之一。Objective：To observe the effect and mechanism of Jianpi Shugan Hewei decoction on expression of gastric emptying and interstitial cells of cajal（ICC） in functional dyspepsia（FD） rats. Methods：FD model was induced by moderately enraging the rats＂ tail. All rats were randomly divided into 4 groups：a control group, a model group, a Chinese herbology group and h western medicine group Drugs were administrated for 14 days. ICC and GE were determined by immunohistochemical method. Gastric emptying was observed with phenolsul- fonphthalein. Results：Compared with the control group, gastric emptying and the expression of ICC in the model group were obviously decreased （P 〈 0.05）. Compared with the model group, gastric emptying and ICC increased significantly in Chinese herbology group and western medicine group（P 〈0.05 ）. Conclusion： Jianpi Shugan Hewei decoction may increase diet and water intake, promote ICC expression in FD rats and accelerate gastric emptying.
