摘 要:目的:了解莪黄汤含药血清对人结肠癌SW480细胞增殖的影响及对Wnt/β-catenin信号通路中β-catenin、cyclin D1蛋白表达的影响。方法:通过莪黄汤不同浓度含药血清处理SW480细胞,MTT法检测细胞增殖,流式细胞技术检测细胞周期,免疫组化及Western blot检测Wnt/β-catenin通路中β-catenin、Cyclin D1蛋白的表达。结果:MTT检测显示不同浓度莪黄汤含药血清在体外对结肠癌SW480细胞增殖有明显抑制作用,呈时间依赖及浓度依赖关系;流式细胞检测显示莪黄汤可抑制SW480细胞增殖,并提示莪黄汤可能在S期发挥作用,阻止细胞进入G2/M期,其中7.14g/kg组抑制作用最明显;免疫组化及Western检测提示莪黄汤可降低β-catenin及其下游cyclin D1蛋白的表达。结论:莪黄汤可能通过Wnt/β-catenin信号通路抑制SW480细胞增殖发挥抗肿瘤作用。Objective: to knowing the effect of Ehuang decoction containing serum on human colon cancer SW480 cell proliferation and the effect on the expression of β-catenin and cyclin D1 protein in Wnt/ β-catenin signaling pathway. Methods: SW480 cells were treated by Ehuang de- coction containing serum with different concentrations; cell proliferation was detected by MTT. ;cell cycle was detected by Flow cytometry ; Wnt β-catenin signaling in β-catenin and cyclin D1 expression was detected by immunohistochemistry and Western blot . Results: MTT detection showed that the proliferation of colon cancer SW480 cells is significantly inhibited by Ehuang decoction(莪黄汤) containing serum with different concentrations in vitro , which in time-dependent and concentration dependent manner; flow cytometry showed that Ehuang de- coction could inhibit the proliferation of SW480 cells, and suggested that Ehuang decoction may play a role in S phase of preventing the ceils from enter into G2 / M phase, the 2G / L group has the most significant inhibition effect (P 〈 0.05 ) ; immunohistochemistry and Western detection suggested that Ehuang decoction 7.14g/kg can reduce the expression of β-catenin and its downstream cyclin D1 protein. Conclu- sion : Ehuang decoction could inhibit the proliferation of SW480 cells by Wnt/ β-catenin signaling pathway, which shows anti-tumor activities.
