血尿安胶囊抗尿路感染的药效学研究

摘　　要：目的：评价血尿安胶囊防治尿路感染的作用,为其临床应用提供实验依据。方法：采用大肠ATCC25922标准菌株制备大鼠膀胱逆行泌尿系统感染模型,血尿安胶囊10g/kg、5g/kg、2.5g/kg连续灌胃给药6d,观察药物对模型大鼠尿常规、血清肌酐与尿素氮含量,膀胱洗液菌落计数的影响,并剖取肾脏进行脏器指数、菌落计数及病理组织学检查。采用琼脂对倍稀释法和纸片法检测血尿安胶囊体外对大肠埃希菌、金黄色葡萄球菌、铜绿假单胞菌、阴沟肠杆菌、变形杆菌等临床分离菌株的抗菌作用。结果：血尿安胶囊显著降低膀胱逆行泌尿系统感染模型大鼠肾脏、膀胱洗液的菌落计数,降低尿蛋白阳性率和血清尿素氮水平,降低大鼠肾脏组织的炎性病变程度,以5g/kg剂量为优。血尿安胶囊体外对泌尿系统易感菌的抑菌范围为0.056~0.1g原生药/ml,对铜绿假单胞菌、变形杆菌有较强杀灭作用,杀菌浓度〈0.45g原生药/ml。结论：血尿安胶囊具有抗尿路感染作用,其作用机理有待进一步探讨。Objective： the prevention and cure effect of Xueniao＇an Capsule on urinary tract infection was evaluated to provide the experimental evi- dence for its efficacy and clinical application. Methods： Eseherichia coli ATCC25922 standard strain was used to establish the bladder retro- grade urinary tract infection rat model, Xueniao＇an Capsule （10g/kg, 5g/kg, and 2.5g/kg, respectively） was administered by orally for 6 days. The effects of drugs on the urine routine, serum ereatinine and urea nitrogen levels, and colony count of the bladder lotion in the blad-der retrograde urinary tract infection rats were observed. In addition, the organ index, colony count, and histopathological examination of the kidney were calculated and performed. The antibacterial effects of Xueniao＇an Capsule on clinical isolated bacterial strains including Esche- richia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter cloacae, Bacillus proteus etc. were assayed by agar dilution meth- od and disk diffusion method in vitro. Results： Xueniao＇an capsule significantly reduced the colony count of kidney and bladder washings in bladder retrograde urinary system infection model rats, urinary protein positive rate and serum urea nitrogen level and inflammatory lesions of renal tissues were reduced. The dose of 5g/kg had better effects than the other two doses. The inhibition range of Xueniao＇an capsule to the susceptible bacteria of urinary system was 0.056 - 0.1 g crude drug/ml in vitro, and Xueniao＇an capsule showed powerful exterminate action to Pseudomonas aeruginosa and Bacillus proteus, and the bactericidal concentration was less than 0.45g/ml. Conclusions ： Xueniao＇an cap- sule has protective effects against urinary tract infection and the mechanisms need further studies.
