摘 要:目的:评价黄芩、南五味子在防治肝纤维化方面所表现的相互作用。方法:将SD雄性大鼠随机分为对照组、模型组、黄芩提取物2g/kg、南五味子浸膏2g/kg以及黄芩提取物2g/kg+南五味子浸膏2g/kg,每天灌胃给药一次,连续4周,对照组和模型组则给以等体积的0.5%CMC;同时,除对照组外,其余各组动物均腹腔注射二甲基亚硝胺(DMN)10mg/kg,每天1次,每周连续3d,持续4周。末次给药后24h,取血清检测总胆红素(T-Bi L)、谷草转氨酶(AST)、谷丙转氨酶(ALT)、碱性磷酸酶(ALP),谷氨酰转移酶(γ-GT)、总蛋白(TP)、白蛋白(ALB)等生化指标。取肝脏称其重量,测定羟脯氨酸(Hy P)含量;取同一叶肝脏分别进行HE、Masson染色观察组织形态变化和胶原堆积情况。结果:DMN大鼠血清TP和ALB显著降低,T-Bil、ALP、γ-GT、AST、ALT以及肝脏组织Hy P水平都显著升高;HE染色显示肝脏纤维化明显,Masson染色显示胶原容积百分比显著增加,与对照组比较有统计学差异。南五味子浸膏2g/kg可显著对抗DMN引起的肝脏指数降低、可显著增加DMN大鼠血清ALB,显著降低DMN大鼠血清T-Bi L、γ-GT、AST和ALT活性。其肝脏纤维化病变以及胶原堆积均明显减轻;黄芩提取物2g/kg可显著对抗DMN肝脏胶原纤维的生成;黄芩提取物2g/kg+南五味子浸膏2g/kg可显著对抗DMN引起的肝脏指数降低,降低DMN大鼠血清T-Bi L、AST、ALT、γ-GT和肝脏组织Hy P,显著改善其肝脏病理组织结构以及胶原堆积。方差分析结果显示南五味子浸膏2g/kg、黄芩提取物2g/kg在增加DMN大鼠血清ALB水平和降低其肝脏组织Hy P方面表现出显著的协同增效作用。结论:研究结果表明黄芩与南五味子配伍能协同防治DMN诱导的大鼠肝纤维化,其协同作用机理值得进一步探讨。Objective: The aim is to evaluate the effect of Radix Scutellaria(RS) and Fructus Schisandrae Sphenanthera(FSS) on liver fibrosis. Methods: SD rats were divided into 5 groups randomly:normal control group, DMN (Dimethylntrosamine), RS 2g/kg, FSS 2g/kg, RS 2g/ kg and FSS 2g,/kg. The rats ig different concentration of drugs, or the same volume of 0.5% CMC-Na, once a day for 4 weeks. Rats were in- duced liver fibrosis by intraperitoneally injection of DMN 10mg/kg once a day during continuous 3 days in a week, for 4 weeks. After 24h of the last administration, the rat serum was taken to measure related biochemical indicators i. e. T-BiL, AST, ALT, ALP, γ-GT, TP and ALB. Meanwhile, the weight and content of Hyp of livers were measured. The liver histomorphology and eonagen accumulation were observed by HE and Masson staining. Results:Compared to the control, the levels of TP and ALB in serum obviously decreased, the levels of T-BiL, ALP, γ-GT, AST, ALT in serum and Hyp in liver tissue were pronouncedly increased, and the liver fibrosis was clearly observed after HE staining, and the ratio of collagenous tissue content in livers was significantly increased in model rats. FSS 2g/kg could obviously suppress the decrease of liver index induced by DMN, significantly increase the level of ALB and decreased the levels of T-BiL, γ-GT, AST and ALT in serum, and decreased fibrosis and collagen accumulation in livers in DMN treated rat. RS 2g/kg could obviously decrease the collagen ac- cumulation induced by DMN. Meanwhile the RS 2g/kg + FSS 2g/kg could significandy lower the level of T-BiL, AST, ALT and γ-GT in serum and HyP content in liver tissue, obviously suppress the decrease of liver index induced by DMN ,decrease the levels of T-BiL,γ-GT, AST and ALT in serum,significantly alleviate liver fibrosis lesions and collagen accumulation in DMN treated rats. FSS 2g/kg and RS 2g/kg exhibited synergistic effect on decreasing the level of collagen accumulation and the content of HyP in liver tissue. Conc
