摘  要:目的建立测定血清万古霉素质量浓度的高效液相色谱法。方法以Agilent HC-C18柱(4.6×150mm,5um)为色谱柱,流动相为甲醇-磷酸二氢钾缓冲液(20∶80),流速为1.0ml/min,检测波长为238nm,以6%高氯酸为沉淀血清蛋白,离心后取上清液直接进样测定。结果万古霉素的低、中、高(2.0,16.0,64.0mg/L)3种浓度的准确度介于98%-110%之间,日内及日间精密度的RSD均小于8.5%(n=5);分析方法的定量下限浓度为2mg/L;线性范围为2.0-64.0mg/L,回归方程为y=13.871x+8.8127(R=0.9996,P<0.05,n=7)。结论该方法灵敏、准确、简单、快速,适用于血清万古霉素血药浓度的监测和药物代谢动力学研究。Objective To estsblish a HPLC method for the determination of serum mass concentration of vancomycin. Methods Vancomycin was analyzed by the RP-HPLC system with Agilent HC-C18 column(4.6×150mm,5um) and the mobile phase of Methanol-potassium dihydrogen phosphate(20 ∶80). The flow rate was 1.0m L/min and the UV detection wavelength was 238 nm.With 10% potassium permanganate as the precipitation serum albumin,the supernatant after centrifugation was taken for detection by direct sample injection. Results The accuracy rate of vancomycin in low,medium and high concentrations(2.0,16.0,64.0mg/L)were all between 98-110%. The inter-day and intra-day relative standard deviations(RSD) were less than 5%(n=5). The lower limits of quantification of serum vancomycin was 2.0mg/L. The linear range was 2.0-64.0mg/L. The regress equations was y =13.871x+8.8127(R2=0.9996,P〈0.05,n=7). Conclusion This method shows to be sensitive,accurate,simple,rapid,and suitable for monitoring the serum concentration of vancomycin and its pharmacokinetic study.
