摘  要:目的探讨过氧化物酶体增殖物激活受体γ2基因C1431T多态性在人群中的分布及其与超重、肥胖症和膳食因素的关系。方法从血凝块中提取DNA,用聚合酶链反应和限制性片段长度多态性方法(PCR-RFLP)检测330例成年超重、肥胖症人群以及227例对照人群的PPARγ2基因C1431T多态性;对研究对象进行体格检查、膳食调查。结果 CC、CT和TT三种基因型的频率分别为32.0%、48.7%和19.3%,符合Hardy-Weinberg定律。超重、肥胖症人群和正常组人群的基因型CC、CT和TT及等位基因C和T构成比差异有统计学意义(χ~2=13.16,P=0.001;χ~2=5.34,P=0.021)。三种基因型人群的总能量、蛋白质、脂肪和碳水化合物摄入量无显著性差异。结论 PPARγ2基因C1431T多态性与超重、肥胖症有关。Objective To study the genotype frequencies of peroxisome proliferators-activated-receptors-gamma2 C1431 T gene and its possible association with the overweight,obesity and dietary intakes. Methods The PCR-PFLP method was used to detect the polymorphism of PPARγ2 C1431 T gene of 330 adults with overweight,obesity and 227 normal adults. Their physical examinations,dietary investigation were analyzed. Results The genotype frequencies of PPARγ2 C1431 T CC,CT and TT were 32.0%,48.7% and 19.3% respectively,which were in agreement with Hardy-Weinberg equilibrium(P〈0.05). There was significant difference in the distribution of genotypes or allele between the overweight,obesity group and the control group(χ~2=13.16,P=0.001;χ~2=5.34,P =0.021). There was no significant difference in total energy、protein、fat and carbohydrate ingestion in three genotypes(P〈0.05). Conclusion The results suggested PPARγ2 gene C1431 T polymorphism associated with the overweight,obesity.
