淫羊藿苷对去卵巢大鼠骨质疏松症的保护作用及其机制

摘　　要：目的:观察淫羊藿苷对去卵巢大鼠骨质疏松症的影响,并探讨其作用机制。方法:50只6月龄雌性大鼠,随机分为假手术组、模型组、阳性药组、低剂量和高剂量淫羊藿苷组,每组10只。阳性药组大鼠每周灌胃给予1mg·kg^(-1)尼尔雌醇;低和高剂量淫羊藿苷组大鼠每天灌胃给予50和100mg·kg^(-1)淫羊藿苷。手术摘除双侧卵巢建立骨质疏松动物模型,术后2周开始给药,给药12周后麻醉状态下腹主动脉取血处死大鼠,观察大鼠骨密度(BMD)、骨组织形态学、血清生化指标和细胞凋亡相关蛋白的变化。结果:与假手术组比较,模型组大鼠BMD降低(P<0.01),血清中钙水平下降(P<0.05),血清磷水平升高(P<0.05),血清骨钙素(BGP)、血清碱性磷酸酶(ALP)、一氧化氮(NO)和一氧化氮合酶(NOS)水平明显降低(P<0.05或P<0.01);与模型组比较,阳性药组和高剂量淫羊藿苷组大鼠BMD明显升高(P<0.01),血清钙水平升高、血清磷水平下降(P<0.05),BGP和ALP水平明显升高(P<0.05)。与假手术组比较,模型组皮质变薄,骨小梁宽度变窄,Bax和caspase-3表达水平上调(P<0.01),Bcl-2表达水平下调(P<0.01);与模型组比较,低和高剂量淫羊藿苷组大鼠可见骨皮质增厚,骨小梁宽度变宽,Bax和caspase-3表达水平下调(P<0.05或P<0.01),Bcl-2表达水平上调(P<0.05或P<0.01)。结论:淫羊藿苷对去卵巢所致的大鼠骨质疏松症具有保护作用,该作用可能与抑制细胞凋亡有关。Objective：To observe the effect of icariin on the osteoporosis of the ovariectomized rats,and to explore the mechanism.Methods：Fifty female rats aged 6 months were randomly divided into sham operation group, model group,positive drug group,low dose of icariin group and high dose of icariin group （n= 10）.The rats in positive drug group were given with 1 mg· kg-1 nilestriol every week;the rats in low and high doses of icariin groups were given with 50 and 100 mg · kg-1 icariin every day.The bilateral ovaries of rats were excised by operation to establish the osteoporosis models.2 weeks after operation,the rats were treated with icariin for 12 weeks,and then they were sacrificed by drawing blood from abdominal aorta under anesthesia condion.The bone mineral density （BMD）,serum biochemical indicators,bone histomorphology and the expressions of apoptosis-related proteins were detected.Results：Compared with sham operation group,the BMD of the rats in model group was decreased （P 〈0.01）,the serum calcium level was decreased （P 〈0.05）,and the serum phosphorus level was increased （P 〈0.05）,and the serum BGP,ALP,NO and NOS levels were significantly decreased （P 〈0.05 or P 〈0.01）.Compared with model group,the BMD of the rats in positive drug group and high dose of icariin group were obviously increased （P 〈 0.01 ）, the serum calcium levels were increased, the phosphorus levels were decreased （P 〈0.05）,and the BGP and ALP levels were increased significantly （P 〈 0.05 ）. Compared with control group,the cortical in model group was thinned,the width of bone trabecula was reduced,the Bax and Caspase-3 expression levels in bone tissue were increased,and the Bcl-2 expression level was decreased.Compared with model group,the cortical in icariin group was thicked,the width of bone trabecula was increased,the Bax and Caspase-3 expression levels were decreased,and the Bcl-2 expression level was increased （P 〈0.05 or P 〈0.01）.
