半乳糖凝集素-3在脓毒症中的作用研究

摘　　要：目的：通过动物实验，研究半乳糖凝集素-3基因缺陷小鼠是否较野生型小鼠对细菌性脓毒血症更为易感。方法1.采用革兰氏阴性鼠伤寒沙门氏菌分别感染年龄和体重相符的野生型小鼠和半乳糖凝集素-3基因缺陷小鼠，监测并比较两型小鼠的存活率；2.评估并比较其对外周循环以及内脏器官包括肝、脾、肺、肾细菌的清除率。3.采用MSD细胞因子多分析系统检测两型小鼠在细菌感染后的不同时间点其血清炎症因子的表达水平。结果与野生型小鼠相比，半乳糖凝集素-3基因缺陷小鼠感染革兰氏阴性鼠伤寒沙门氏菌后存活率降低，其外周循环的细菌清除能力受损，血清炎症因子的表达水平上升。结论与野生型小鼠相比，半乳糖凝集素-3基因缺陷小鼠对细菌性脓毒症更为易感。Objective To determine whether gal3-deficient mice are more susceptible to microbial sepsis than the wild-type littermates through the experiments on animals.Methods 1.Infecting the gal3-deifcient and the wild-type mice whose ages and weight are nearly equivalent with G- salmonella typhimurium, monitoring and comparing the survival rate of the two types mice. 2.Evaluating and comparing the clearance rate of bacteria in their peripheral circulation and visceral organs including liver, spleen, lung, kidneys. 3.Testing the inflammatory factors＇ expression level in serum of the two type mice by MSD system.Results Compared with the wild-type mice, the survival rate of the gal3-deifcient mice infected with G- salmonella typhimurium drop down, the clearance rate of bacteria in peripheral circulation is damaged, and the inflammatory factors＇ expression level rises. Conclusion Compared with the wild-type mice, the gal3-deifcient mice are more susceptible to microbial sepsis.
