摘 要:目的对家族性拇外翻相关基因进行初步定位,为中医治疗拇外翻提供理论依据。方法通过流行病学调查,确定该病遗传因素群体的诊断指标,收集目前已知样本量最大的一个足拇趾外翻家系样本,对其进行全基因组扫描和连锁分析,并对这些区域包含的基因的特点进行分析。结果找到6个相关的染色体区域,多是与骨骼和肌肉发育相关的基因,这些基因的表达会受到益肾填精、补钙壮骨中药复方的影响。结论本研究结果为进一步研究足拇趾外翻分子遗传机理提供最直观的依据,也为中医诊治拇外翻提供基因分子水平的佐证。Objective To make initial location of familial hallux valgus relating genes and to provide theoretical basis for Chinese medicine treatment of hallux valgus. Methods By epidemiological investigation,the diagnostic criteria of the disease genetic factor group were confirmed. A family sample of hallux valgus was collected with the biggest sample size known so far. Whole-genome scan and linkage analysis was made on the sample. The characteristics of genes contained in these regions were analyzed. Results Six relating chromosomal regions were found; most were genes related to bones and muscles development. The expressions of these genes could be influenced by Chinese herbal compound of invigorating kidney and semen,calcium supplement and strengthening bones. Conclusion The research result might provide the most intuitive basis for further study on hallux valgus molecular genetic mechanism. It might also provide evidence of gene molecular level for Chinese medicine treatment of hallux valgus.
