拉米夫定对禽白血病病毒的体内外抑制效应及其应用

摘　　要：为鉴定逆转录酶抑制剂类药物拉米夫定(LAM)对禽白血病病毒(ALV)体内和体外抑制效应及其在辅助ALV净化中的作用,本研究首先通过在培养液中添加不同浓度LAM连续进行DF-1细胞传代,并利用CCK-8法测定细胞活性,确定添加不超过5μg/m L的LAM对于DF-1细胞生长无影响,但与不添加药物的对照组相比可以显著抑制J亚群ALV(ALV-J)在DF-1细胞中的增殖。体内试验证实,在4 mg/只/d连续用药7 d情况下,感染ALV-J鸡群在第9周时病毒血症完全消除。此外,新城疫病毒活疫苗人工污染A亚群ALV(ALV-A)后,利用LAM连续干预,结果显示,经过LAM两代干预后可有效去除毒种中ALV-A污染。结果表明LAM对ALV-J和ALV-A均具有抑制作用,有助于降低ALV感染率和疫苗外源病毒污染,从而促进不同鸡群中ALV的净化。The purpose of this study is to identify and investigate the inhibitory effects of Lamivudine （LAM） on avian leukosis virus （ALV） both in vivo and in vitro. In the present study, the DF-1 cells were passaged with different concentrations of LAM, and cell activity was detected by a cell counting Kit-8 （CCK-8）. In vitro treatment group, the LAM had no effect on DF-1 cells with a concentrations less than 5 μg/mL, on the contrary, the replication of ALV-J on DF-1 cells was inhibited significantly compared to control group. In vivo treatment group, the results showed that ALV-J viremia was completely eradicated at 9 week-old in chickens, which were inoculated with ALV-J and treated continuously for 4 mg/chicken/day with LAM at 7 day-old. Moreover, the ALV-A contamination in Newcastle disease virus （NDV） stock strains could be eradicate after two passages in cell cultures with LAM. In conclusion, these results demonstrated that LAM had the ability to inhibit the replication of ALV-J and ALV-A both in vitro and in vivo. Therefore, the LAM could be used to eradicate ALV-J in chicken flocks, reducing the ALV infection and contamination in vaccine stock virus strain.
