摘  要:目的:观察肺积方对Lewis肺癌移植小鼠肿瘤生长的抑制作用,并探讨其可能的作用机制。方法:30只近交系C57BL/6纯系小鼠行Lewis肺癌细胞移植造模后随机分为模型组、肺积方组及环磷酰胺(CTX)化疗组,每组10只。模型组只行造模,肺积方组予每日0.4 ml(含生药0.8 g)灌胃给药;CTX组按15 mg/kg溶于0.2 ml生理盐水中腹腔注射,每周1次;各组均常规喂食。30 d后观察记录各组小鼠瘤体大小;观察期满后各组小鼠断颈处死,留取肿瘤组织进行CD4^+CD25^+Treg流式细胞检测。结果:CTX组及肺积方组肿瘤相对体积(RTV)均较模型组缩小(P<0.05),肺积方组RTV低于CTX组(P<0.05);各组相对肿瘤体积抑制率比较,肺积方组高于CTX组(P<0.05);各组肿瘤组织中CD4^+CD25^+Treg比例比较,肺积方组及CTX组均较模型组降低(P<0.05),CTX组与肺积方组比较差异无统计学意义(P>0.05)。结论:肺积方有抑制Lewis肺癌移植小鼠肿瘤生长的作用,其抑制作用可能与减少CD4^+CD25^+Treg细胞分化有关。Objective: To observe the inhibition effect of Feijifang on tumor growth in mice transplanted with Lewis lung cancer and discuss its possible mechanism. Methods: Thirty inbred C57BL/6 mice transplanted with Lewis lung cancer were randomly divided into the model group, Feijifang group and cyclophosphamide chemotherapy group ( CTX group ), 10 in each group. The model group were received any treatment. The mice in Feijifang group were intragastrically administrated with Feijifang decoction at a dose of 0.4 ml a day( including 0.8 g crude drug). CTX were intraperitoneally injected into the CTX group mice at a dose of 15 mg/kg once a week. Conventional feeding was conducted in all group. After 30 days, the tumor size was observed and recorded in each group. Then the mice were sacrificed and the tumor tissues were collected and the CD4+ CD25 + Treg cell were detected by flow cytometry. Results : The relative tumor volume (RTV) in the CTX and Feijifang group was decreased compared with the model group ( P 〈 0.05 ), and the RTV in the Feijifang group was lower than that in the CTX group ( P 〈 0.05 ) ; the inhibition rate of RTV of the Feijifang group was higher than that of the CTX group ( P 〈 0.05 ) ; the ratio of CD4 + CD25 + Treg in the Feijifang group and CTX group were decreased compared with the model group ( P 〈 0.05 ), but there was no statistical difference between the Feijffang group and CTX group (P 〉0.05). Conclusion: Feijifang decoction could inhibit the tumor growth in mice transplanted with Lewis lung cancer, and the mechanism may be related to suppressing the differentiation of CD4 + CD25 + Treg.
