摘 要:目的从自杀相关因子及其配体(Fas/Fas L)角度探讨消瘿合剂对自身免疫性甲状腺炎的作用机制。方法 SD大鼠58只,除正常组10只大鼠外,其余48只大鼠建立实验性自身免疫性甲状腺炎(EAT)大鼠模型,随机分为模型组、雷公藤多苷对照组和消瘿合剂低剂量、高剂量组。采用放免法(RIA)检测血清甲状腺球蛋白抗体(Tg Ab)水平;免疫组化法检测甲状腺上皮细胞Fas、Fas L表达。结果与正常组比较,模型组Tg Ab升高(P<0.05);与模型组比较,雷公藤多苷组、消瘿合剂高剂量组能显著降低Tg Ab(P<0.05)。与正常组比较,模型组Fas、Fas L表达阳性面积差异均有统计学意义(P<0.01);与模型组比较,干预各组均能使Fas、Fas L表达降低,差异均有统计学意义(P<0.05,P<0.01)。在降低Fas表达方面,消瘿合剂高剂量组、低剂量组与雷公藤多苷组比较,差异有统计学意义(P<0.01);在降低Fas L表达方面,与雷公藤多苷组比较,消瘿合剂高剂量组差异无统计学意义(P>0.05),消瘿合剂低剂量组差异有统计学意义(P<0.01)。结论消瘿合剂治疗自身免疫性甲状腺炎的细胞凋亡机制可能是通过降低甲状腺上皮细胞Fas、Fas L表达达到的。Objective To explore the mechanism of Xiaoying Mixture on autoimmune thyroiditis in view of factor associated suicide / factor associated suicide ligand ( Fas/FasL). Methods For 58 rats, 10 rats were divided into the normal group, and rat model of experimental autoimmune thyroiditis was established in the other 48 rats. Then the successful modeling rats were divided into the model group ,tripterygium glycosides group as positive control and Xiaoying Mixture groups with low- and high-does. The level of serum thyroid globulin antibody (TgAb) was detected by radioimmunoassay (RIA) ; and the expression of Fas and FasL in thyroid epithelial cells were detected by immunohistochemistry. Results Compared with the normal group, the level of TgAb in the model group was increased (P 〈0.05 );compared with the model group, the level of TgAb in the tripterygium glycosides group and Xiaoying Mixture group with high-dose was significant decreased(P 〈0.05 ). The difference on the positive area of Fas and FasL expression between the normal group and model group was statistically significant ( P 〈0.01 ) ; compared with the model group, the expression of Fas and FasL in the treatment groups was decreased with statistically significant difference (P 〈0.05, P 〈0.01 ). For the effect on reducing the expression of Fas, the difference between the tripterygium glycosides group and Xiaoying Mixture groups with low- and high-dose was statistically significant (P 〈0.01 ). For the effect on reducing the expression of FasL, the difference between the tripterygium glycosides group and Xiaoying Mixture group with high-dose was not statistically significant ( P 〉 0.05 ) ; the difference between the tripterygium glycosides group and Xiaoying mixture group with low-dose was statistically significant ( P 〈0.01 ). Conelusion The mechanism of Xiaoying Mixture on cell apoptosis in the treatment of autoimmune thyroiditis may be related to reducing the expressions of Fas and FasL in thyroid epitheli
