锦鲤疱疹病毒减毒株的筛选及免疫原性研究

摘　　要：为了致弱锦鲤疱疹病毒得到减毒株,试验采用细叶桉叶的提取液与病毒在细胞中共培养来致弱病毒,并以中和抗体试验检验弱毒株的免疫原性。结果表明:利用获得的减毒株在加强免疫42天时,抗锦鲤疱疹病毒中和抗体效价达到最高水平,同时灭活病毒免疫组的抗锦鲤疱疹病毒中和抗体水平低于KHV-P_(52)免疫组,PBS对照组未检测出抗锦鲤疱疹病毒的中和抗体,两个免疫组与对照组比较差异显著(P<0.05);KHV-P52免疫组对鲤鱼的免疫保护效果好,免疫保护率为100%。说明细叶桉叶提取液能有效致弱锦鲤疱疹病毒,通过试验获得了一株锦鲤疱疹病毒弱毒株。To attenuate Koi herpesvirus （KHV） to obtain an attenuated strain of KHV, the extracting solution of Eucalyptus tereticornis leaves and KHV were co - cultured in the cells to attenuate the virus in the test, and the immunogenicity of the attenuated strain was tested by neutrali- zing antibody test. The results showed that the neutralizing antibody titer against koi herpesvirus reached the highest level using the attenuated strain of KHV at 42 d of strengthening immunity, meanwhile, the level of neutralizing antibody against KHV in the group immunized with inacti- vated virus was lower than that in the group immunized with KHV - Ps2, and the neutralizing antibody against KHV was not detected in PBS control group. The difference was significant （ P 〈 0.05 ） between the two immune groups compared with the control group. The protective effect of the group immunized with KHV -P52 on common carp was good clue to its 100% immune protection rate. The results indicate that the extracting solution of Eucalyptus tereticornis leaves can effectively attenuate KHV, and an attenuated strain of KHV is obtained through the test.
